华蟾素对人脑胶质瘤细胞SHG44增殖的影响

摘  要:目的探讨华蟾素对人脑胶质瘤细胞SHG44增殖、凋亡的影响及其机制。方法不同浓度华蟾素分别作用于SHG44细胞株,MTT法及流式细胞技术检测其对SHG44细胞株增殖的影响和对细胞周期的影响;(AO/EB)双重染色及透射电镜观察SHG44细胞株形态学变化和超微结构的改变;Westernblot技术检测Caspase8蛋白及Bcl-2的表达。结果1μg/ml和10μg/ml浓度下的华蟾素显著抑制人脑胶质瘤细胞sHG44增殖作用(P〈0.05),并呈浓度和时间依赖性。FCM检测发现华蟾素可使人脑胶质瘤细胞SHG44的G0/G1期细胞明显增多(P〈0.05),可见凋亡峰;Western Blot检测显示华蟾素作用可使Caspase 8蛋白表达上调,Bcl-2蛋白表达下调。结论华蟾素能够诱导Caspase-8和Bcl-2的差异性表达,具有抑制人脑胶质瘤细胞SHG44的细胞增殖,促进其凋亡的作用。Objective To explore the influence of cinobufotalin on the proliferation and apoptosis of human glioma cell line SHG44 and its mechanism. Methods Different concentrations cinobufotalin were applied to SHG44 cell line, MTT assay cinobufacini on SHG44 cell line proliferation; flow cytometry cinobufacini SHG44 cell line for cell cycle; TEM ultrastructural changes SHG44 cell plant type, ( AO / EB ) double staining morphological changes; Westernblot Caspase8 to detect Bel-2 protein and the expression. Results Cinobufotalin ( 1 Ix g/ml and 10 Ix g/ml concentration ) inhibited human glioma cells SHG44 proliferation ( P〈0.05 ) , and in a dose and time dependent. FCM detected that cinobufotalin make human glioma cells SHG44 the GO / (31 phase ceils increased significantly ( P〈0.05 ) , and can see the apoptotic peak; Western Blot test showed cinobufotalin role in bringing Caspase8 protein upregulation, Bcl-2 protein was down-regulated. Conclusion Cinobufotalin can induce differential expression of caspase 8 and Bcl-2, and play the role in inhibiting proliferation of tumor cells and accelerating apoptosis of SHG44 cells.
