摘 要:目的观察银杏叶提取物(ginkgo biloba extract,Egb)对血管平滑肌细胞(vascular smooth muscle cell,VSMC)凋亡的影响,初步探讨其可能机制。方法采用组织贴块法培养胎儿主动脉VSMC。在不同Egb浓度及培养时间下,检测各组细胞凋亡率,并检测Fas及Bcl-2的表达量。结果凋亡细胞比率结果显示,Egb组凋亡细胞比率明显增加,在浓度为800μg/mL时间为24 h组的凋亡细胞比率最高,而且最高值已经达到了35.50%±0.63%;Fas蛋白阳性细胞率与Egb浓度和时间的增加呈现一定的正相关。Fas蛋白阳性细胞率在6h组为5.67%±1.04%,24 h组增加为11.00%±0.56%左右,Egb在浓度为800μg/mL时间为24 h组比率最高达到33.20%±1.22%;Bcl-2蛋白阳性细胞率与Egb浓度和作用时间呈现明显的负相关,Egb组在浓度为800μg/mL时间为24 h组比率降低到8.33%±0.87%。结论 Egb可以诱导VSMC的凋亡;Egb诱导VSMC的凋亡过程与Fas表达增加和Bcl-2的表达减少有着直接的联系。Objectives To observe the effect of ginkgo biloba extract (Egb) on vascular smooth muscle cell (VSMC) apoptosis, and to preliminary discuss its possible mechanism. Methods Fetal aortic VSMC was cultured by tissue piece holding method. Cell apoptosis rate in different incubation duration and concentration of Egb was tested. Fas and expression of Bcl-2 were also detected. Results Ratio of apoptotic cells in Egb group significantly increased, the highest ratio was in concentration of 800μg/mL with incubation duration of 24 h group which reached 35.50%±0.63%. Fas protein positive-cell rate was positively correlated with concentration and incubation duration of Egb. Rate of Fas protein positive cell in 6 h group was 5.67%±1.04%, in 24 h group increased to 11.00%±0.56%; the highest rate of Fas protein positive cell was in concentration of 800μg/mL with duration of 24 h group which reached to 33.20%±1.22%. Bcl-2 protein positive-cell rate was negatively correlated with Egb concentration and incubation duration, the lowest Bcl-2 protein positive-cell rate was in Egb concentration of 800 μg/mL with incubation duration of 24 h group which reached to 8.33%±0.87%. Conclusions Egb can induce the apoptosis of VSMC, which is directly correlated with the increasing expression of Fas and decreasing expression of Bcl-2.
