摘 要:目的探讨大肠杆菌疫苗对发育期胎兔肺表面活性物质磷脂生成的影响。方法将孕兔30只随机分为3组,大肠杆菌疫苗组10只,地塞米松组10只,生理盐水组10只。大肠杆菌疫苗组和生理盐水组分别于妊娠第10、17、24天宫颈肌肉内注射大肠杆菌疫苗0.1 ml/kg和生理盐水0.1 ml/kg,地塞米松组于妊娠第22、23、24天分别肌肉注射地塞米松0.4 ml/kg。于妊振第27天剖宫产,行支气管肺泡灌洗收集管肺泡灌洗液(BALF),采用高效液相色谱-蒸发光散射法检测BALF中二棕榈磷脂酰胆碱(DPPC)的浓度,采用二喹啉甲酸(BCA)法测定BALF上清液总蛋白浓度,采用动态定时散射比浊法测定羊水和BALF中CRP浓度;采用免疫荧光法测定羊水和BALF中PCT浓度。采用单因素方差分析;LSD和Duunet T3法进行统计学分析。结果地塞米松组、大肠杆菌疫苗组、生理盐水组胎肺BALF中DPPC的浓度分别为(44.89±0.29)μg/ml、(44.62±0.11)μg/ml、(40.20±0.27)μg/ml,地塞米松组和大肠杆菌疫苗组胎肺BALF中DPPC的浓度明显高于生理盐水组,差异有统计学意义(P<0.01);大肠杆菌疫苗组、地塞米松组、生理盐水组BALF上清液总蛋白浓度分别为(0.409±0.012)μg/ml、(0.253±0.043)μg/ml、(0.244±0.019)μg/ml,大肠杆菌疫苗组BALF上清液总蛋白浓度高于地塞米松组和生理盐水组,差异有统计学意义(P<0.01);大肠杆菌疫苗组羊水和BALF中CRP、PCT明显高于地塞米松组和生理盐水组,差异有统计学意义(P<0.01)。结论大肠杆菌疫苗诱导宫内感染和产前注射地塞米松对胎兔肺DPPC生成均有明显的促进作用,接种大肠杆菌疫苗可能对胎肺上皮细胞造成一定程度的损伤。Objective To investigate the effect of E. coli vaccine on the generation of surfactant phospholipids in fetal rabbit lungs. Methods Thirty pregnant rabbits were randomly divided into three groups, ten rabbits in E. coli vaccine group, ten rabbits in dexam- ethasone group, and ten rabbits in normal saline group. In E. coli vaccine group and normal saline group, rabbits were given E. coli vaccine or saline 0. lml/kg through intramuscular injection of cervix on Day 10, Day 17 and Day 24 during gestation, and in dexamethasone group, rabbits were given dexamethasone 0.4ml/kg intramuscularly on Day 22, Day 23 and Day 24 during gestation. All the rabbits received cesare- an delivery on Day 27 of gestation, and bronchoalveolar lavage fluid (BALF) specimens were collected using bronchoalveolar lavage, and the DPPC concentrations of surfactant phospholipids in BALF were determined quantitatively by high performance liquid chromatography - evapo- rative light scattering, meanwhile, total protein concentrations in BALF supernatant were determined using bicinchoninic acid (BCA) meth- od, the CRP concentrations in BALF supernatant and amniotic fluid were determined using dynamic timing nephelometry; the PCT concentra- tions in BALF supernatant and amniotic fluid were determined using immunofluorescence. One-way ANOVA, LSD and Dunnet T3 methods were applied for statistical analysis. Results The DPPC concentrations of BALF in fetal lungs in dexamethasone group, E. coli vaccine group and normal saline group were respectively (44. 89±0.29) μg/ml, (44. 62±0. 11) μg,/ml, (40. 20±0.27)μg/mL The DPPC con- centrations in dexamethasone group and E. coli vaccine group were higher than that in normal saline group with significant differences (P〈 0.
