摘 要:[目的]本文旨在探讨IGF-Ⅰ和MSTN基因在鸭胚胸肌成肌细胞的表达规律。[方法]以生长速度不同的高邮鸭和金定鸭为试验对象,采用实时荧光定量PCR方法检测鸭13、15、17、19、21和23胚龄时胸肌成肌细胞IGF-Ⅰ、MSTN mRNA的表达情况,并进行品种间比较。[结果]IGF-Ⅰ与MSTN mRNA表达都呈先升后降的趋势,都存在17胚龄的表达高峰,19胚龄之后,表达显著下调并持续至23胚龄;高邮鸭IGF-ⅠmRNA的表达比金定鸭多了13胚龄的表达高峰,且在13胚龄时,2个基因的表达量都是高邮鸭极显著高于金定鸭(P<0.01)。与之相反,IGF-Ⅰ/MSTN mRNA表达量的比值都是呈先降后升的趋势,在19胚龄之前比值小于1,21胚龄至23胚龄时,表达量比值显著升高;品种间是高邮鸭的显著高于金定鸭(P<0.05)。IGF-Ⅰ与MSTN mRNA表达均无显著品种效应,而IGF-Ⅰ/MSTN mRNA表达量比值品种效应显著。2个品种的IGF-Ⅰ与MSTN mRNA表达均呈极显著正相关。[结论]IGF-Ⅰ与MSTN协同调节成肌细胞的增殖与分化,二者表达量比值在品种上的差异可能是造成鸭胚胸肌发育品种差异的重要因素。[Objectives]The present experiment was conducted to investigate the development expression patterns of IGF-Ⅰ and MSTN genes in duck pectoral muscle myoblasts. [Methods]Fluorescent real-time quantitative PCR was applied to detect the distribution profile of IGF-Ⅰ and MSTN mRNA in pectoral muscle myoblasts of Gaoyou ducks and Jinding ducks at the embryonic days 13,15,17,19,21 and 23. [Results]The results showed that the expression of IGF-Ⅰ and MSTN mRNA increased gradually between embryonic days 13 and 17,peaked at embryonic days 17,then declined gradually until embryonic days 23. The embryonic days 13 was an expression peak of IGF-Ⅰ mRNA observed in Gaoyou duck but not in Jingding duck. The expression of IGF-Ⅰ and MSTN in Gaoyou duck was higher than that of Jinding duck on embryonic days 13( P 0. 01). In contrast,the expression of IGF-Ⅰ to MSTN ratio was less than 1 before embryonic days 19,and increased significantly from embryonic days 21 to 23. Besides,the ratio in Gaoyou duck was higher than that of Jinding duck. There was no significant variety effect was found in the expression of IGF-Ⅰ and MSTN,but found significant variety effect in the expression of IGF-Ⅰ to MSTN ratio. The expression of IGF-Ⅰ was positively correlation with that of MSTN. [Conclusions]The results showed that the balance between proliferation and differentiation of pectoral muscle myoblast in duck embryos were jointly regulated by IGF-Ⅰ and MSTN. The varietal differences on the ratio of IGF-Ⅰ to MSTN maybe play a key role on duck embryo pectoral muscle development.
