摘　　要：细菌感染是引起奶牛乳腺炎的主要原因,而胞壁酰二肽(MDP)几乎存在于所有细菌中,核苷酸结合寡聚化结构域2(NOD2)受体是天然免疫中对病原微生物的病原体相关分子模式识别的一类感受器,在机体天然免疫应答中发挥重要作用。为了研究MDP对小鼠乳腺组织和乳腺细胞NOD2表达的影响,试验以小鼠为模型,采用不同浓度的MDP在体乳腺内灌注和对体外小鼠乳腺上皮细胞用不同浓度的MDP刺激,观察小鼠乳腺组织的病理组织学变化,乳腺组织及细胞中NOD2、下游的受体作用蛋白2(RIP2)表达的变化,以及炎性细胞因子TNF-α、IL-6的表达变化。结果表明:从乳腺灌注不同浓度MDP后可引起急性乳腺炎,乳腺组织中炎性细胞增多;MDP刺激后乳腺组织及细胞中NOD2、RIP2、TNF-αmRNA表达水平明显升高,细胞IL-6 mRNA表达水平没有明显升高。说明NOD2介导信号途径促使炎性细胞因子的表达。The bacterial infection is caused mainly due to mastitis in dairy cows, while muramyl dipeptide （MDP） exists in almost all bacteria. Nucleotide - binding oligomerization domain 2 （NOD2） receptors are a class of receptors in the innate immune system, which recognize patho- gen - associated molecular pattern and play an important role in the body＇ s innate immune response. To study the effect of MDP on NOD2 ex- pression in mammary tissue and mammary epithelial cells of mice, mice were used as model in the test. Different concentrations of MDP were used to perfuse the mammary glands of mice, and were also used to stimulate the mammary epithelial cells of mice in vitro. Histopathological changes were observed in the mammary tissue of mice, and changes in the expressions of NOD2 in the mammary tissue and cells, receptor inter- acting protein - 2 （ RIP2 ） in the downstream, as well as the inflammatory cytokines TNF - α and IL - 6 were also observed. The results showed it caused acute mastitis after perfusing different concentrations of MDP in the mammary glands of mice, and inflammatory ceils were increased in the mammary tissue. Expression levels of NOD2, RIP2, TNF -α in the mammary tissue and cells were significantly increased after stimulation with different concentrations of MDP, but the expression level of IL - 6 mRNA levels in the cells was not significantly increased. The results in- dicate that NOD2 - mediated signal pathway induces the expressions of inflammatory cytokines.
