摘 要:目的探索环境温度对左旋多巴体外探针回收率的影响程度。方法通过改变灌流液温度与左旋多巴标准溶液的温度,将微透析探针的环境温度分为3种状态:1室温状态:灌流液及左旋多巴标准液均为室温(约24℃);2常规状态:灌流液为室温,标准液温度为体温(约37℃);3体温状态:灌流液及标准液均为体温。分别在3种状态下进行左旋多巴探针回收率测定,采用高效液相色谱电化学系统进行分析,分别计算左旋多巴的体外探针回收率。结果在室温状态下为探针对左旋多巴的回收率为(17.5±1.73)%,在常规状态下为(28.3±2.01)%,在体温状态为(45.4±4.82)%,不同环境温度组的LD探针回收率具有显著性差异(P<0.01)。结论环境温度可显著影响体外探针回收率的测定结果。OBJECTIVE To investigate the effect of environment temperature on the recovery of microdialysis probe for levodopa( LD). METHODS Levodopa was dialyzed with microdialysis probe in different environment temperature. There were three conditions: 1Lower temperature condition,the temperature of LD standard solution and that of the perfusate were both 24℃; 2Normal temperature condition,the temperature of LD standard solution was37℃ and that of the perfusate was 24℃; 3 Higher temperature condition,the temperature of LD standard solution and that of the perfusate were both 37℃. The concentration of LD in the dialyzed solution and the standard solution were analyzed by HPLC-ECD,and recoveries were calculated. RESULTS In lower temperature condition,the recovery of microdialysis probe was( 17. 5 ± 1. 73) %,while in normal temperature condition,that was( 28. 3 ±2. 01) %,in higher temperature condition,that was( 45. 4 ± 4. 82) %. In different temperature conditions,the recoveries for LD were significantly different( P 〈 0. 05). CONCLUSION The environmental temperature of microdialysis probe affects the recovery for LD.
