摘  要:目的观察肉苁蓉苷对C17.2神经干细胞增殖的影响,并探讨其可能的作用机制。方法体外常规培养C17.2神经干细胞,按1∶3传代。传代培养的神经干细胞C17.2,分为给药组0.0625mg·m L-1、0.125mg·m L-1、0.25mg·m L-1和空白组,培养24h。镜下观察C17.2神经干细胞的形态,噻唑蓝(MTT)法和Brdu免疫标记检测细胞的活性及增殖能力。结果 MTT检测结果显示,药物干预后,各组神经干细胞的活性均有增加,与空白组比较,差异有统计学意义(P<0.05);Brdu免疫标记显示,与对照组相比,高中剂量组的Brdu阳性细胞数显著增多。结论肉苁蓉苷可在一定程度上促进神经干细胞的增殖,可能成为神经康复的潜在药物。OBJECTIVE To observe the effects of glycosides of cistanche on proliferation of C17. 2 neural stem cells,and to explore its possible mechanism. METHODS C17. 2 neural stem cells were cultured conventional in vitro and passed at ratio of 1 ∶ 3. Then the cultured C17. 2 neural stem cells were randomly divided into medicated group( 0. 0625,0. 125,0. 25 mg · m L- 1) and control group,and continued to been cultured for 24 hours. The morphology of C17. 2 neural stem cells were observed under microscope,the activity and proliferation of neural stem cells were measured by MTT and Brdu respectively. RESULTS MTT result showed that,compared with the control group,the activity of the neural stem cells in each group were increased after drug intervention,the difference was statistically significant( P 〈 0. 0). Brdu immunofluorescence showed,compared with the control group,the number of Brdu positive cells of high and medium does group increased significantly. CONCLUTION Glycosides of cistanche can promote the proliferation of neural stem cells to a certain extent,can be a potential drugs for nerve recovery.
