摘  要:为了探讨小花棘豆对家兔卵巢α-甘露糖苷酶(AMA)的影响,进一步揭示小花棘豆的毒性作用机理。将24只雌性家兔随机分为4组,即对照组和试验Ⅰ、Ⅱ、Ⅲ组。将小花棘豆全草粉碎后,按Ⅰ组添加15%(含苦马豆素30 mg/kg)、Ⅱ组添加30%(含苦马豆素60 mg/kg)、Ⅲ组添加45%(含苦马豆素90 mg/kg)的比例制作混合饲料,饲喂至典型临床症状出现为止。攻毒后第14、35、70天每次每组随机采集2只家兔的卵巢,检测家兔卵巢AMA活性及表达的变化。试验组及对照组家兔卵巢高尔基体α-甘露糖苷酶Ⅱ(AMA1)和溶酶体α-甘露糖苷酶(AMA2)均有表达,但各试验组表达转录水平与对照组差异显著,试验Ⅰ组家兔AMA1、AMA2以及试验Ⅱ组AMA2的表达与对照组差异不显著(P﹥0.05),试验Ⅱ组AMA1以及试验Ⅲ组家兔AMA1、AMA2的表达均显著低于对照(P﹤0.05),第70天时试验Ⅲ组家兔AMA1活性和表达均极显著低于对照(P﹤0.01)。小花棘豆中毒可影响家兔卵巢AMA的活性及其基因的转录表达。To investigate the effects of content and transcriptional expression of α-mannosidase( AMA) in ovary tissue poisoning by Oxytropis glabra DC,and reveal the toxicity mechanism of O. glabra DC in future,24 female rabbits were randomly and equally divided into 4 groups( the control group and experimental groupⅠ,Ⅱ,Ⅲ),the dried plant of O. glabra DC was smashed,and different amounts of the grass powder( 15 %,30 % and 45 % the corresponding swainsonine content was 30,60,90 mg / kg) were mixed with the feeds in the three experimental group. The rabbits consumed the forages freely until typical symptoms were observed. 2 rabbits were selected randomly from each group on the 14 th,35th and 70 th day respectively,the ovary was collected and the contents of AMA and its mRNA expression in different encephalic regions were measured. It was found that AMA in golgi and lysosomal expressed in ovary of all experimental groups,O. glabra DC inhibited the expression of ovary AMA1 and AMA2 mRNA,and the difference was obvious. The difference was not obvious in experimental groupⅠ,AMA2 of groupⅡand control group( P ﹥ 0. 05),the expression of AMA1 and AMA2 mRNA decreased in experimental group Ⅲ and AMA1 mRNA decreased in experimental groupⅡ was significant lower than control group( P ﹤ 0. 05),the content and mRNA decreased of AMA1 in experimental group Ⅲ was very significantly lower than control group( P ﹤ 0. 01). The results showed that O. glabra DC has remarkable effects on content and transcriptional expression of AMA in rabbit ovary,in a time-effect as a dose-effect relationship.
