Optimization of Selected Parameters Affecting Selenium Content in Extracts from Agrocybe cylindracea and Anti-fatigue Activity of Extracts

摘  要:根据Box-Behnken的中心组合实验设计原理,在单因素试验的基础上采用3因素3水平的响应面分析法,优化柱状田头菇(Agrocybe cylindracea)硒提取物的提取工艺条件。单因素试验结果表明:适宜浸提次数、提取时间、碱液(KOH)浓度、料液比(g/mL)和浸提温度分别为2次、7h、0.075 mol/L、1∶30和60℃。响应面试验获得的最佳提取工艺:碱液浓度0.073mol/L,提取时间6.91h,料液比1.0∶24.9,提取物中硒浓度预测为22.82g/kg,实际测定值为22.53g/kg。小鼠负重游泳试验结果表明,灌胃硒提取物显著延长小鼠负重游泳时间:低剂量组小鼠的负重游泳时间显著增加(P<0.05),中、高剂量组的负重游泳时间极显著增加(P<0.01)。Single factorial experiments and response surface methodology were used to optimize selected parameters affecting selenium content in extracts of Agrocybe cylindracea fruiting bodies. Optimal values of extraction frequency, extraction time, KOH concentration, solid-liquid ratio (fruit body powder : KOH, g/mL) and extraction temperature were determined by single factorial experiments to be two extractions, 7 h, 0. 075 mol/L, 1 : 30 and 60 ℃. Box-Behnken testing and response surface methodology using the polynomial model Y = 22.42-0.98 A-1.05 B + 1.88 C-1.07 AB + 1.62 AC + 1.02 BC-2.31 A2-3. 43 B2-2, 52 C2 predicted that maximal selenium content in extracts would be achieved with two extractions using 0. 073 mol/L KOH, and a solid:liquid ratio of 1.0:24.9 combined with a extraction time of 6.91 h. Under these optimal condition, the selenium content in extracts was 22. 53 g/kg fruiting bodies. Administration of medium (25 mL/kg-d) and high (75 mL/kg-d) doses of selenium extracts to male mice for 30 days significantly (0.01 level) prolonged their exhaustive swimming time compared with a control group given 25 mL/kg, d distilled water, whereas low doses of selenium extracts significantly prolonged the exhaustive swimming time at the 0.05 level.
