摘 要:【目的】从生理生化角度揭示大豆抗豆卷叶螟机制,为进一步挖掘大豆抗豆卷叶螟基因、创制抗性种质及实现大豆高效抗豆卷叶螟分子育种奠定基础。【方法】以对豆卷叶螟高抗大豆品种赶泰-2-2和高感品种皖82-178为研究对象,分别于人工接上豆卷叶螟4龄幼虫0、6和24 h取样测定并分析其叶片中相关营养物质含量、保护性酶活性、次生物质及植物激素含量。【结果】豆卷叶螟取食前后高抗和高感大豆叶片中的可溶性蛋白和单宁含量变化没有规律。豆卷叶螟取食后,大豆叶片的可溶性糖含量、保护性酶类[超氧化物岐化酶(SOD)、过氧化氢酶(CAT)和多酚氧化酶(PPO)]活性、植物激素(乙烯、茉莉酸和脱落酸)含量均明显增加,且高抗品种赶泰-2-2的SOD活性、乙烯和脱落酸含量总体水平高于高感品种皖82-178。【结论】大豆叶片内可溶性糖含量、CAT和PPO活性及茉莉酸含量的变化可能与豆卷叶螟的诱导有关,与而抗性基因型关系不明显;SOD活性、乙烯和脱落酸含量不仅与豆卷叶螟诱导有关,还可能与大豆抗豆卷叶螟基因有关。[Objective]The mechanism of soybean's resistance to Lamprosema indicata (Fabricius) was investigated from the perspective of physiology and biochemistry, in order to lay foundation for further studying L. indicata (Fabricius)-resis- rant gene of soybean, breeding insect-resistant germplasm and realizing molecular breeding of high-resistant soybean. [Method]The leaves of two soybean cultivars viz., Gantai-2-2 and Wan 82-178 were inoculated with 4th-instar L. indicate (Fabricius) larvae. After 0, 6 and 24 h, different leaves were collected, respectively. Then the nutrient content, protective enzyme activity, secondary biomass and plant hormone content in leaves were determined, respectively. [Result]The re- sults showed that, there was no change rule of soluble protein and tannin contents to be found in leaves of high resistant and susceptible soybean before and after being inoculated with L. indicate(Fabricius) larvae. Furthermore, after being in- oculated with L. indicate (Fabricius) larvae, the soluble sugar content, protective enzymes (SOD, CAT, PPO) activities, plant hormones(ethylene, jasmonic acid, abscisic acid) contents increased significantly, and the overall level of SOD ac- tivity and abscisic acid content in leaves of Gantai-2-2 were higher than that in leaves of Wan 82-i78. [Conclusion]The chances of soluble sugar content, CAT and PPO activities and jasmonic acid content in soybean leaves may be correlated with induction of L. indicate (Fabricius), but have little correlation with resistant genotypes. Moreover, the SOD activity, ethylene and abscisic acid contents be not only correlated with induction of L. indicate(Fabricius), but also correlated with L. indicata(Fabricius)-resistant gene of soybean.
