摘  要:为研究四倍体矮秆小麦拔节期赤霉素(GA)合成途径中涉及的关键酶基因及其与植株矮化的关系,选择拔节期的矮杆番麦和四倍体矮杆小麦近等基因系ANW 16D、ANW 16F、ANW 16G(分别含有Rht 14、Rht 16、Rht 18基因)以及作为对照的高秆小麦Langdon(LD)进行试验。取拔节期小麦穗下第一茎节节间(约0.5cm),提取总RNA并反转录,对影响GA合成过程中的GA 20ox、CPS、GA 2ox、KAO、KO、KS、GA 3ox、GID 1、GA-MYB、XET、GIP、GAI、RSG和14-3-3等关键酶基因进行实时荧光定量分析。结果显示,GA 20ox、GA 2ox、KAO、KO、KS、GA 3ox、GID 1、GA-MYB、XET、GIP、GAI、RSG在4个矮杆品系和高杆对照中均有表达。GA 20ox在高秆小麦中的表达量高于矮秆小麦,GA-MYB与之具有相似的表达模式,说明GA 20ox和GA-MYB基因的低表达影响了矮秆小麦拔节期赤霉素的合成,从而导致矮化性状。To explore the key enzyme gene cause gibberellin (GA) synthesis insufficient which bring about plant dwarf in the synthesis way of GA of tetraploid dwarf wheat in the wheat jointing stage. Five cultivars,namely,aiganfanmai, the tetraploid wheat near isogenic lines of ANW 16 D, ANW 16 F and ANW 16 G, contains Rht 14, Rht 16 and Rht 18 gene, respectively, were served as materials in present studies.The high stalk wheat Langdon(LD)as control. Take internodes part under spike of materials,extract the total RNA and reverse transcription, pick up the key enzyme genes, including GA 20 ox, CPS, GA 2 ox, KAO, KO, KS, GA 3 ox, GID 1, GA-MYB, XET, GIP, GAI, RSG, 14-3-3, which involved in the synthesis way of GA biosynthesis, use realtime-PCR to test transcription level. Results showed that,GA 20 ox, GA 2 ox, KAO, KO, KS, GA 3 ox, GID 1, GA-MYB, XET, GIP, GAI and RSG were expressed in five wheat at jointing stage internodes part.The GA 20 ox and GA-MYB had the similar expression profile in high stalk and dwarf wheat,namely,the expression in high stalk wheat slightly higher than the dwarf wheat.This results indicated that the expression of GA 20 ox and GA-MYB in jointing stage have certain effect in a wheat GA synthesis pathway.The expression of GA 20 ox in dwarf is lower than high stalk control wheat, show that low expression of GA 20 ox and GA-MYB influence the synthesis way of GA which cause the dwarf character.
