摘  要:[目的]从抗氧化生理响应的角度探讨氨氮对萼花臂尾轮虫的毒理机制。[方法]测定萼花臂尾轮虫24 h、48 h及96 h的氨氮半致死浓度(LC50);通过测定萼花臂尾轮虫体内的H_2O_2、丙二醛(MDA)含量和超氧化物歧化酶(SOD)、过氧化氢酶(CAT)活性的变化,研究其对氨氮胁迫浓度和时间的抗氧化生理响应。[结果]轮虫24 h LC50、48 h LC50和96 h LC50的氨氮浓度分别为12.3、6.7和2.3 mg/L。当氨氮浓度达到2.5 mg/L时,轮虫体内24 h内H_2O_2和MDA含量均显著上升;当氨氮浓度达到1.5 mg/L时,轮虫体内24 h内SOD活性显著下降,而当氨氮浓度达到10 mg/L时CAT活性显著下降。在12.3 mg/L氨氮的胁迫下,轮虫SOD活性和CAT活性分别在12 h和24 h显著下降,H_2O_2和MDA含量均在12 h显著升高。[结论]SOD活性和H_2O_2、MDA含量可作为检测氨氮对萼花臂尾轮虫急性毒性的灵敏指标。[Objective] To discuss the toxicological mechanism of ammonia nitrogen to Brachionus calyciflorus from the aspect of antioxidant physiological response. [Method]24 h,48 h and 96 h LC50 of ammonia nitrogen to B. calyciflorus was determined. The changes of H2O2 and malondialdehyde( MDA) contents,activities of superoxide dismutase( SOD) and catalase( CAT) in B. calyciflorus were determined. The antioxidant physiological response of B. calyciflorus to ammonia nitrogen concentration and stress time was studied. [Result] The concentrations of ammonia nitrogen for 24 h LC50,48 h LC50 and 96 h LC50 to B. calyciflorus were 12. 3,6. 7 and 2. 3 mg / L,respectively. When the concentration of ammonia nitrogen reached 2. 5 mg / L,H2O2 content and MDA content in B. calyciflorus within 24 h were significantly increased. When the concentration of ammonia nitrogen was 1. 5 mg / L,SOD activity of B. calyciflorus within 24 h was significantly decreased.When the concentration of ammonia nitrogen was 10 mg / L,CAT activity was significantly decreased. Under the stress of 12. 3 mg / L ammonia nitrogen,SOD activity and CAT activity of B. calyciflorus within 12 h and 24 h significantly decreased,while H2O2 content and MDA content were significantly increased within 12 h. [Conclusion] SOD activity,H2O2 content,and MDA content can be used as sensitive indicators for detecting the acute toxicity of ammonia nitrogen to B. calyciflorus.
