摘 要:目的:研究重组人促红细胞生成素(rHuEPO)对感染引起的肝肾损伤以及内质网应激的保护效果。方法:将雄性Wistar大鼠随机分为阴性对照组、脂多糖(LPS)组和LPS+rHuEPO组(n=30)。LPS组和LPS+rHuEPO组采用尾静脉注射LPS的方法建立感染模型;LPS+rHuEPO组采用尾静脉注射rHuEPO的方法进行干预。干预前和干预后6、12h时,采集血清检测TNF-α、iNOS、BUN、Cr、ALT、AST水平;干预后12h时,采集肝肾组织,检测内质网应激分子(GRP78、CHOP、Caspase-12)的mRNA水平。结果:建模后、rHuEPO干预前,LPS组和LPS+rHuEPO组大鼠血清中TNF-α、iNOS、BUN、Cr、ALT、AST水平均高于阴性对照组,但两组间无显著差异;干预后6、12h时,LPS+rHuEPO组大鼠血清中TNF-α、iNOS、BUN、Cr、ALT、AST水平均低于LPS组;干预后12h时,LPS+rHuEPO组大鼠肝、肾组织中GRP78、CHOP、Caspase-12的mRNA水平低于LPS组。结论:大鼠LPS感染模型中,rHuEPO对感染所致肝肾损伤和内质网应激具有保护作用。Objective: To study the protective effect of recombinant human erythropoietin on liver and kidney injury as well as endoplasmic retieulum stress caused by infection. Methods: Male Wistar rats were randomly divided into negative con- trol group, LPS group and LPS4-rHuEPO group, LPS group and LPS rHuEPO group received injection of LPS via caudal vein to establish infection models, and LPS--rHuEPO group received injection of rHuEPO via caudal vein for intervention; be- fore intervention as well as 6 h and 12 h after intervention, serum was collected to detect TNF-α, iNOS, BUN, Cr, ALT and AST contents; 12 h after intervention, liver and kidney tissue was collected to detect mRNA contents of endoplasmic reticulum stress molecules (GRP78, CHOP and Caspase-12). Results: After model establishment and before rHuEPO intervention, ser- um TNF-α, iNOS, BUN, Cr, ALT and AST contents of LPS group and LPSrHuEPO group had no differences and were higher than those of negative control group; 6h and 12 h after intervention, serum TNF-α, iNOS, BUN, Cr, ALT and AST contents of LPS-4-rHuEPO group were lower than those of LPS group; 12 h after intervention, mRNA contents of GRP78,CHOP and Caspase-12 in liver and kidney tissue of LPS rHuEPO group were lower than those of LPS group. Conclusion: Recombinant human erythropoietin has protective effect on liver and kidney injury as well as endoplasmic reticulum stress caused by infection.
