摘  要:目的研究二甲基亚砜(DMSO)对酵母多糖诱导的大鼠肾脏损伤的保护作用。方法将清洁级SD大鼠分为4组:SS组、SD组、ZS组及ZD组,每组各30只。ZS组和ZD组大鼠于腹腔注射750 mg/kg酵母多糖以制备肾损伤模型,SS组和SD组大鼠仅予以假手术操作。而后SD组与ZD组大鼠于1 h后皮下注射3 ml/kg DMSO,SS组与ZS组予以等剂量生理盐水。于造模后4、8、24 h分批处死大鼠,每批10只。采用激光多普勒血流仪测定各时间点肾血流量,并抽取腹主动脉血检测肌酐水平,取肾组织测定其含水率、丙二醛(MDA)及髓过氧化物酶(MPO)活性。结果造模后4、8、24 h,与SS组和SD组比较,ZS和ZD组肾血流量显著降低,肾组织含水率、MDA含量、MPO活性及血肌酐含量均显著升高(P均<0.05)。且与ZS组比较,ZD组造模后4、8、24 h肾血流量明显升高[(119±13)BPU vs.(156±22)BPU;(103±11)BPU vs.(121±12)BPU;(87±11)BPU vs.(108±11)BPU],肾MDA含量显著降低[(6.2±0.6)nmol/mg Pro vs.(5.3±0.4)nmol/mg Pro;(9.1±0.9)nmol/mg Pro vs.(5.6±0.5)nmol/mg Pro;(10.7±1.0)nmol/mg Pro vs.(6.7±0.6)nmol/mg Pro]。而ZD组肾含水率仅在8、24 h时[(75.7±2.2)%vs.(72.4±2.7)%;(79.2±2.3)%vs.(74.9±2.2)%],血肌酐含量仅在24 h时[(61.4±8.6)U/L vs.(36.5±6.7)U/L]较ZS组显著降低(P均<0.05)。但两组大鼠肾MPO活性在各时间点比较,差异均无统计学意义(P均>0.05)。结论DMSO对酵母多糖引起的肾缺血和氧自由基损伤有显著的保护作用。Objective To study the protective effects of dimethyl sulfoxide(DMSO)against renal injury induced by zymosan in rats. Methods SD rats were randomly divided into4 groups: group SS, group SD, group ZS and group ZD, 30 rats in each group. Rats in group ZS and group ZD were given 750 mg / kg zymosan by intraperitoneal injection to prepare renal injury model, rats in group SS and group SD only performed sham-operation. And rats in group SD and group ZD were received 3 ml / kg DMSO by subcutaneous injection on 1 h after building the model, and rats in group SS and group ZS were received equivalent dose of normal saline at the same time. The rats were further divided into 4, 8, 24 h subgroup after building the model, 10 rats in each subgroup. The blood flow of kidney was monitored with laser-Doppler flow meter, the plasma creatinine, malondialdehyde(MDA), myeloperoxidase(MPO) and water content from specimens of kidney were detected at 4, 8, 24 h after building the model. Results Compared with group SS and group SD, the blood flow of kidney were significantly lower, and the content of water and MDA, the activity of MPO and plasma creatinine were significantly higher in group ZS and group ZD at 4, 8, 24 h after building the model(allP〈0.05), and the blood flow of kidney [(119 ± 13) BPU vs.(156 ± 22) BPU;(103 ± 11) BPU vs.(121 ± 12) BPU;(87 ± 11) BPU vs.(108 ± 11) BPU] were lowest, the content of MDA [(6.2 ±0.6) nmol / mg Pro vs.(5.3 ± 0.4) nmol / mg Pro;(9.1 ± 0.9) nmol / mg Pro vs.(5.6 ± 0.5) nmol / mg Pro;(10.7 ±1.0) nmol / mg Pro vs.(6.7 ± 0.6) nmol / mg Pro] were highest in group ZD. However, the water content of kidney only at 8, 24 h [(75.7 ± 2.2)% vs.(72.4 ± 2.7)%;(79.2 ± 2.3)% vs.(74.9 ±2.2)%] and the plasma creatinine only at 24 h [(61.4 ± 8.6) U / L vs.(36.5 ± 6.7)U / L] in group ZD were significantly lower than those in group ZS(allP〈0.05).
