摘  要:目的观察晚期炎性因子高迁移率族蛋白B1(HMGBl)在诱导大鼠胰腺腺泡细胞Janus激酶2/信号转导与转录激活子3(JAK2/STAT3)信号通路活化中的作用。方法取胰腺分离胰腺腺泡细胞,随机分为A组:正常对照组、B组:HMGB1促进组(终质量浓度1mg/L)、C组:JAK2抑制剂-AG490处理组(25μmol/L)、D组:STAT3抑制剂-雷帕霉素处理组(40μg/L),采用实时定量反转录·聚合酶链反应(RT-qPCR)和Western blot法检测10、30、60、120min时间点细胞中JAK2、STAT3mRNA表达及60、120min时间点JAK2、STAT3蛋白表达水平。结果与A组比较,B组各时间点JAK2 mRNA(3.5221±0.3112、5.5446±1.2479、28.4939±2.0742、12.5023±0.5025)及30、60、120min时间点STAT3 mRNA的相对表达量升高(10.5726±1.1449、44.8684±2.0218、7.2113±1.4239),60、120min时间点JAK2(0.3441±0.0414、0.4872±0.0184)、STAT3蛋白表达升高(0.6380±0.0109、0.3861±0.0121),差异有统计学意义(P〈0.01);与B组比较,c、D组各个时间点JAK2 mRNA的相对表达量(2.1207±0.1025、3.7891±0.5083、13.3262±1.4876、3.7323±0.6125;2.7262±0.4991、2.3952±0.3466、9.6233.±1.0955、4.8082±0.6442)及30、60、120min时间点STAT3 mRNA的相对表达量下降(5.8233±1.1793、11.6400±0.8085、2.6956±0.4514;3.0244±0.5343、14.9727±0.1877、5.4984±0.3878),差异有统计学意义(P〈0.05);60、120min JAK2(0.2587±0.0141、0.3654±0.0103;0.2702±0.0558、0.3874±0.0135)、STAT3蛋白相对表达量(0.4567±0.0179、0.3272±0.0184;0.4777±0.0178、0.3509±0.0070)显著降低(P〈0.01)。结论HMGB1可诱导大鼠胰腺腺泡细胞JAK2/STAT3信号通路的活化。Objective To research whether the Janus kinase 2/signal transducer and activator of transcription 3 (JAK2/STAT3) signaling pathway activated by highly mobility group box- 1 (HMGB1) in inducing rat pancreatic acinar cells. Methods The rat pancreaticacinar cells were isolated, and randomly divided into four group: A group: normal controlgroup, B group: HMGB1 promotion (final concentration 1 g/L), C group: AG490 treatmentgroup (25 txmol/L)and D group: RPM treatmentgroup (40 μg/L). Real - time quantitative reverse transcription polymerase chain reaction ( RT - qPCR) and Western blotting detected the expression of JAK2, STAT3 mRNA at the time of 10, 30, 60, 120 min and the protein expression of JAK2, STAT3 at the time of 60, 120 rain. Results Compared with group A, the relative expression levels of the JAK2 mRNA of group B ( 3. 522 1 ± 0. 311 2, 5. 544 6 ± 1. 247 9, 28. 493 9 ± 2. 074 2, 12. 502 3 ±0. 502 5) at each time point and the STAT3 mRNA of group B at 30, 60, 120 min time point were increased ( 10. 572 6 ± 1. 144 9, 44. 868 4 ±2. 021 8, 7. 211 3 ± 1. 423 9), the protein expression of JAK2 (0.344 ± 0.0414, 0.4872s 0.0184) and STAT3 (0.6380± 0.0109, 0. 386 1 ± 0. 012 1 ) at 60, 120 min time point was elevated.
