摘  要:目的观察大豆苷元(DAI)对动脉粥样硬化(As)的抑制作用,并探讨其机制。方法ApoE^-/-小鼠分为两组(n=8):对照组高脂喂食8周后行生理盐水灌胃6周,实验组高脂喂食8周后行大豆苷元(5.0mg/kg)灌胃6周。灌胃完成后油红O染色比较两组小鼠腹大动脉根部斑块面积;酶联免疫吸附试验(ELISA)检测各组小鼠血清脂质指标和炎性因子浓度;免疫蛋白印迹分析动脉内皮细胞中黏附分子表达量变化。结果灌胃6周后,对照组斑块面积为(0.28±0.04)mm^2,实验组为(0.144.0.02)mm^2;ELISA实验结果中,对照组总胆固醇(TC)、三酰甘油(TG)和低密度脂蛋白胆固醇(LDL—C)的水平分别为(10.3±0.9)、(3.2±0.3)、(8.6±0.7)mmol/L,实验组分别为(8.2±0.8)、(2.1±0.4)、(6.0±0.8)mmol/L;对照组白细胞介素(IL)-6、IL-1、肿瘤坏死因子(TNF)-α和单核细胞趋化蛋白(MCP)-1的浓度分别为(32.1±4.6)、(85.3±10.2)、(153.6±18.7)、(635.9±72.5)ng/L,实验组浓度分别为(21.3±3.4)、(40.3±6.9)、(95.8±12.3)、(403.6±55.6)ng/L;Western blot分析实验中,对照组血管内皮细胞黏附分子(VCAM)-1、细胞间黏附分子(ICAM)-1和E选择素(E-selectin)相对表达水平分别为1.26±0.18、1.03±0.12、1.62±0.21,实验组分别为0.46±0.06、0.30±0.04、0.67±0.10。结论大豆苷元通过抑制炎性反应和黏附分子表达减轻动脉粥样硬化。Objective To study the inhibitory effect of Daidzein on atherosclerosis, and to explore the molecular mechanism. Methods ApoE^ -/- mice were divided into two groups ( n = 8 each) : control group and experimental group. The mice in control group were fed on a high fat diet for 8 weeks and then received gavage with normal saline each day for additional 6 weeks. The mice in experimental group were fed on a high fat diet for 8 weeks and received gavage with Daidzein (5.0 mg/kg) each day for additional 6 weeks. Plaque areas in the root of the abdominal aorta were measured using oil - red O staining. Serum lipid parameters and inflammatory factors were detected with specific enzyme linked immunosorbent assay (ELISA) kits. The adhesion molecules in vascular endothelial cells were determined using Western blotting analysis. Results After garage for 6 weeks, plaque areas in the control group and the experimental group were (0. 28 ±0. 04) and (0. 14 ±0.02) mm^2, respectively. ELISA showed that the levels of total cholesterol (TC), TG and LDL -C in the control group and the experimental group were (10. 3 ±0.9), (3.2 ± 0. 3) and (8. 6 ±0. 7) vs. (8.2 ±0. 8), (2. 1 ±0. 4) and (6. 0 ±0. 8) retool/L, respectively. The concentrations of interleukin (IL) - 6, IL - 1, tumor necrosis factor - α( TNF - α) and monocyte chemotaetic protein - 1 ( MCP - 1 ) in the control group and the experimental group were ( 32. 1 ± 4. 6), ( 85.3 ± 10.2), (153.6 ± 18.7), (635.9 ±72.5) vs. (21.3 ±3.4), (40.3 ±6.9), (95.8 ± 12.3), (403.6 ±55.6) ng/L, respectively. The relative expression of VCAM - 1, ICAM - 1 and E - seleetin in the control group and the experimental group was 1.26 ± 0. 18, 1.03 ± 0. 12, 1.62 ± 0. 21 vs. 0. 46 ± 0. 06, 0. 30 ± 0. 04, 0. 67 ± 0. 10, respectively. Conclusion Daidzein alleviates atherosclerosis through inhibiting inflammatory response and the expression of adhesion molecules.
