摘 要:目的探讨钙蛋白酶抑制剂ALLN(n—acetyl-Leu—Leu—Norleucinal)与外源性泛素-蛋白酶体片段Ⅱ(UPFⅡ)单用及联用对去血清饥饿大鼠骨骼肌细胞凋亡的影响及机制。方法离体培养3~5日龄SD大鼠骨骼肌细胞,苏木素一伊红(HE)染色及骨骼肌肌动蛋白免疫荧光方法鉴定骨骼肌细胞,传至5~6代骨骼肌细胞稳定成熟,随机分为5组进行干预:A组(完全培养基对照组)、B组(去血清饥饿对照组)、C组(去血清饥饿+UPFⅡ组)、D组(去血清饥饿+UPFⅡ+ALLN组)、E组(去血清饥饿+ALLN组)。药物干预72h后,噻唑蓝(MTT)法测定各组细胞的存活率,实时荧光定量聚合酶链反应(FQ-PCR)检测各组细胞中钙蛋白酶(Calpain)-3、肌肉环指蛋白-1(MuRF-1)、Atrogin-1的mRNA表达。结果MTT结果显示:与A组比较,B、C、D、E组细胞存活率分别为(57.89±5.07)%、(41.22±4.87)%、(56.94±6.02)%、(69.72±6.25)%,存活率均下降(P〈0.05);FQ-PCR结果显示:与A组比较,B、C、D、E组Calpain-3、MuRF-1、Atrogin-1的mRNA表达量分别为23.71、23.18、22.22倍;24.52、24.10、23.29倍;23.78、23.11、22.15倍;22.60、22.37、21.65倍,差异有统计学意义(P〈0.05);E组细胞存活率较B、C、D组均高(P〈0.05),各基因mRNA表达量均明显降低(P〈0.05);D组细胞存活率较C组高(P〈0.05),各基因mRNA表达量均明显降低(P〈0.05);B组与D组之间差异无统计学意义(P〉0.05)。结论适量ALLN在一定程度上能有效降低去血清饥饿诱导大鼠骨骼肌细胞的凋亡率;适量ALLN在一定程度上可以减弱UPFⅡ对去血清饥饿大鼠骨骼肌细胞凋亡的促进作用。Objective To study the effects of Calpain inhibitor N - acetyl - Leu - Leu - Norleucinal (ALLN) and ubiquitin/proteasome fractionⅡ( UPF Ⅱ ) alone or in combination on apoptosis of pri- macy cultured SD rat skeletal muscle cells. Methods The skeletal muscle cells of 3 - 5 - day old SD rats were cultured in vitro and identified by hematoxylin and eosin (HE) staining and skeletal muscle actin ( α - sarcometric actin) immunofluorescence. The skeletal muscle cells were mature and stable after five or six generations of passage, and then were randomly divided into 5 groups : ( 1 ) complete medium : group A (complete medium control group) ; (2) basal medium: group B (serum starvation control group), group C ( serum starvation + UPF Ⅱ ), group D ( serum starvation + UPF Ⅱ + ALLN ), group E ( serum starvation + ALLN). Seventy - two h after drug intervention, the surval rate of skeletal muscle cells was determined by methyl thiazol tetrazolium (MTT) assay and the mRNA expression of Calpain - 3, Muscle ringfinger - 1 ( MuRF - 1 ) and Atrogin - 1 in each group was detected by real - time fluorescent real - time fluorescent quantitative polymerase chain reaction (FQ - PCR). Results The MTT results showed that cell survival rates in groups B, C, D and E was respectively (57.89 ±5.07) %, (41.22 ±4. 87 ) %, (56. 94 ±6. 02 ) %, and ( 69. 72 ±6. 25 ) %, which was significantly lower than in group A ( P 〈 0. 05 ). FQ - PCR revealed that the mRNA expression of Calpain - 3, MuRF - 1, and Atrogin - 1 in groups B, C, D and E was respectively 23.71/23. 18/22. 22, 24. 52/24. 10/23.29, 23.78/23.11/22. 15, 22. 60/22. 37/21.65 times higher than in group A (P 〈 0. 05 for all). The cell survival rate in group E was significantly higher (P 〈 0.05 ) and the expression of Calpain - 3, MuRF - 1, and Atrogin - 1 mRNA was significantly lower ( P 〈 0. 05 ) than groups B, C and D.
