摘 要:目的观察腺病毒携带骨形成蛋白7(BMP-7)基因对小鼠膝关节假体松动模型假体周围组织炎性反应的影响。方法120只8周龄无特定病原体(SPF)级BABL/c雌性小鼠随机分为空白对照组、阳性对照组和BMP-7干预组,构建膝关节假体松动模型。检测小鼠假体周围组织BMP-7 mRNA和核因子(NF)-κB mRNA表达,并测定炎性因子水平。结果与空白对照组和阳性对照组比较,BMPG干预组T2-T4时小鼠假体周围组织中BMP-7 mRNA相对表达量(3.61±0.29、5.03±0.41和6.14±0.38)均升高(P〈0.05);与空白对照组和阳性对照组比较,BMPM干预组T1~T4时小鼠假体周围组织中白细胞介素(IL)-1β[(134.7±9.8)、(153.5±14.2)、(169.4±12.7)、(178.6±11.3)ng/L]、IL-6[(66.2±8.4)、(77.6±7.3)、(91.4±15.2)、(107.2±11.5)ng/L]、肿瘤坏死因子(TNF)-α[(79.5±10.6)、(96.2±12.4)、(118.3±14.1)、(133.7±13.2)ng/L]和NF—κB mRNA(1.59±0.22、2.05±0.19、2.67±0.25和3.04±0.23)均降低(P〈0.05)。结论腺病毒携带BMPG基因可在小鼠膝关节假体松动模型假体周围组织内表达,可抑制假体周围炎性因子的释放。Objective To investigate the effects of recombinant adenovirus carrying bone morphogenetic protein 7 ( BMP - 7) gene on inflammatory response in periprosthetic tissue of mice knee prosthesis loosening model. Methods 120 cases of 8 week - old specific pathogen free (SPF) BABL/c female mice were randomly divided into control group, positive control group and BMP - 7 intervention group ( n = 40 each). Knee prosthesis loosening models were constructed. The mRNA expression of BMP- 7 and nuclear factor - κB (NF - κB) was detected. The inflammatory cytokine levels were measured. Results Compared with the control group and the positive control group, the relative expression levels of BMP - 7 mRNA (3.61 ±0. 29, 5.03 ±0. 41 and 6. 14 ±0. 38) in the BMP -7 intervention group at T2 -T4 were increased (P 〈 0. 05 ). Compared with the control group and the positive control group, the levels of interleukin (IL) -1β [(134.7 ±9.8), (153.5 ± 14.2), (169.4 ± 12.7), (178.6 ± 11.3) ng/L], IL -6 [(66.2±8.4), (77.6 ±7.3), (91.4 ± 15.2), (107.2 ± 11.5) ng/L], tumor necrosis factor (TNF) -α [(79.5 ±10.6), (96.2 ±12.4), (118.3 ±14.1), (133.7 ±13.2) ng/L] andNF-κB mRNA ( 1.59 ± 0. 22, 2. 05 ± 0. 19, 2. 67 ± 0. 25, 3.04 ± 0. 23 ) of periprosthetic tissues at T2 - T4 in the BMP - 7 intervention group were significantly decreased ( P 〈 0. 05 ). Conclusion Adenoviral - mediated BMP - 7 gene could be expressed in the periprosthetic tissues of the mice knee prosthesis loosening model. It could inhibit the inflammatory eytokine release around the prosthesis.
