摘  要:目的利用环孢素构建免疫抑制小鼠,将人乳腺癌细胞接种于小鼠背部,建立一种新小鼠乳腺癌模型。方法40只雌性BALB/c小鼠,在注射细胞前3d,每只小鼠注射环孢素30mg/(kg·d),随机平均分成4组,按照注射人乳腺癌MDA—MB-231细胞量多少分为对照组、低细胞量组、中细胞量组、高细胞量组。将人乳腺癌细胞接种于小鼠背部,观察小鼠成瘤时间、成瘤率、肿瘤PET-CT影像及病理学表现等。结果高细胞量组小鼠成瘤率最高(80%),但小鼠死亡率也最高(40%);中细胞量组小鼠成瘤率次之(70%),但与高细胞量组比较,差异无统计学意义(P〉0.05),但小鼠死亡率(10%)较高细胞量组明显降低;低细胞量组小鼠成瘤率(20%),与前两组相比明显降低,此组实验过程中无小鼠死亡;对照组无小鼠死亡,亦无小鼠成瘤。但对于成像方面的研究,低细胞量组的细胞数量则比较适合。结论利用环孢素成功构建人乳腺癌非裸鼠移植新模型,此模型可以更好地研究免疫反应对肿瘤的影响。Objective In order to establish a new mouse model of breast cancer by injecting the human breast cancer cell into the dorsum of immunosuppressed male which was achieved via a daily intraperitoneal injection of cyclosporine 30 mg/( kg· d). Methods Forty female BALB/c mice were randomly divided into four groups, with 10 in each group. Experimental groups of rats were divided into control group, low cells group, medium cells group, high cells group. The mice were observed in terms of the time of tumorigenesis, rate of the tumor formation, PET - CT imaging and pathology. Results In the group of high cells group, the mortality rate of the mice is the highest in the four groups (80%), moreover, the rate of tumor formation is higher as well (40%). By contrast, refer to the group with low cells group, it states completely opposite result. Compare to the high cells group, the rate of tumor formation in the medium cells group is less (70%), but there is an apparent drop - off in the mice mortality rate ( 10% ). It was showed that there were no death and tumor formation in control group. Conclusion Human breast carcinoma model in mice can be successfully established by this method. By using this model, the process of immune response to the tumor can be much better studied.
