摘 要:目的:研究GPR30在子痫前期胎盘组织和正常胎盘组织中的差异表达及其临床意义。方法:收集子痫前期胎盘组织和正常胎盘组织,检测GPR30的表达量;培养人脐静脉内皮细胞,分别采用GRP30抑制剂、GPR30激动剂联合缺氧复氧进行处理,检测细胞凋亡情况以及促血管新生分子、凋亡分子的含量。结果:子痫前期胎盘组织中GRP30的mRNA含量和蛋白含量显著低于正常胎盘组织;G15处理组的细胞凋亡率显著高于对照组,上清液中VEGF、bFGF的含量显著低于对照组,细胞中Bax、Caspase-3、Caspase-9的mRNA含量显著高于对照组;H/R组的凋亡率显著高于对照组,上清液中VEGF、bFGF的含量显著低于对照组,细胞中Bax、Caspase-3、Caspase-9的mRNA含量显著高于对照组;G1处理组的凋亡率显著低于H/R组,上清液中VEGF、bFGF的含量显著高于H/R组,细胞中Bax、Caspase-3、Caspase-9的mRNA含量显著低于H/R组。结论:胎盘组织中GPR30低表达与子痫前期的发生密切相关,增强GPR的功能能够减少内皮细胞凋亡、增加促血管新生因子的含量,具有内皮保护效应。Objective: To study the differential expression of GPR30 in preeclampsia placenta tissue and normal placenta tissue and its clinical significance. Methods: Preeclampsia placenta tissue and normal placenta tissue were collected and GPR30 expression levels were deteeted human umbilical vein endothelial cells were cultured and processed with GRP30 inhibitor and GRP30 agonist combined with hypoxia-reoxygenation respectively, and cell apoptosis as well as pro-angiogenesis molecule and apoptosis molecule contents were detected. Results: mRNA content and protein content of GRP30 in preeclampsia placenta tissue were significantly lower than those in normal placenta tissue apoptosis rate of G15 group was significantly higher than that of control group. VEGF and bFGF contents in supernatant were significantly lower than those of control group, and mRNA contents of Bax, Caspase-3 and Caspase-9 in cells were significantly higher than those of control group. Apoptosis rate of H/R group was significantly higher than that of control group, VEGF and bFGF contents in supernatant were significantly lower than those of control group, and mRNA contents of Bax, Caspase-3 and Caspase-9 in cells were significantly higher than those of control group. Apoptosis rate of G1 group was significantly lower than that of H/R group, VEGF and bFGF contents in supernatant were significantly higher than those of H/R group, and mRNA contents of Bax, Caspase-3 and Caspase-9 in cells were significantly lower than those of H/R group. Conclusion: Low expression of GPR30 in placenta tissue is closely associated with the occurrence of preeclampsia. EnhancedGPR function can reduce endothelial cell apoptosis and increase the contents of pro-angiogenesis factors. It has endothelial protection effect.
