A SIMPLE AND TIME-SAVING METHOD FOR THE PREPARATION OF PORCINE GROWTH HORMONE

摘  要:将猪垂体在酸性(pH5.5)条件下制备垂体匀浆液,经硫酸铵分级分离后,用Scphadex G-100凝胶柱层析纯化,得到电泳纯的分子量为21.7KD的猪生长激素,产率为1.22g/kg新鲜垂体。胫骨法测定具有促生长活性,其氨基酸组成、氮末端氨基酸及电荷不均一等性质均与文献报道基本一致。此法较之前人的方法有简便易行,周期短的优越性,活性组分集中,得率也较高,是制备有活性的猪生长激素较为理想的方法。We purified porcine Growth Hormone (pGH) by making homogenate of pig pituitary glands at pH 5.5, followed by fractionation with ammonium sulfate and chromatography on Sephadex G-100 column. The purified fraction has a molecular weight of 21.7KD and shows biological activity in the tibia test. Its amino acid composition, N-terminal amino acid as well as properties on electrophoresis are the same as reported in the literatures. The overall yield is about 1.22 grams per kilogram fresh pituitary glands. As compared to other methods reported, it is simple, time-saving and has relatively high yield. We consider that it.is an ideal method for the purification of pGH from pig pituitary glands.
