摘  要:目的探讨大鼠腹主动脉移植后,血管平滑肌细胞(VSMC)的分离、培养方法。方法建立BN至Lewis腹主动脉移植的慢性排斥反应、动脉硬化模型,60d后用组织块贴壁方法分离、培养血管平滑肌细胞并进行传代。采用形态学和肌动蛋白免疫组化鉴定证实为VSMC。结果细胞呈典型的“峰.谷”样生长,历次传代鉴定均为高纯度VSMC,传代至6代左右仍能保持原有性状,细胞活力未见减低。结论组织块贴壁法培养大鼠腹主动脉移植后vsMc具有操作简单、细胞活力高、纯度高等优点,可作为研究器官移植慢性排斥反应、移植物动脉硬化新的体外实验模型。Abstract To explore a method for isolating and culturing vascular smooth muscle cells ( VSMC ) after rat abdominal aortic transplantation. Methods The chronic rejection model of abdominal aortic transplantation was established using BN rats as donors and Lewis rats as recipients. Two months later, the allograft was harvested and VSMCs were isolated and cultured by explanting culture method. VSMC were identified by morphology and the immunohistochemical SABC method ( α -SMActin ) . Results The cultured VSMC showed typical "peak-valley" pattern and kept high purity and original activity after nearly 6 passages. Conclusion Tissue culture is a simple method for isolating and culturing VSMC after rat abdominal aortic transplantation, and the VSMC can remain high activity and high purity. It may provide a new model to study the chronic rejection and allograft arteriosclerosis in organ transplantation in vitro.
