摘  要:目的研究环氧化酶-2在甲状腺乳突状癌、桥本氏甲状腺炎及非毒性甲状腺肿中的表达及作用。方法选取90例甲状腺疾病患者,应用分子生物学分析法从细针抽吸活组织检查和术后组织取材检查提取RNA,酶联免疫吸附测定分别分析c DNA反转录和COX-2基因表达。结果甲状腺乳头状癌中COX-2 mRNA表达明显高于桥本氏甲状腺炎和非毒性结节性甲状腺(P<0.05);甲状腺乳头状癌COX-2表达水平明显高于非毒性结节性甲状腺肿组(P<0.05);细针抽吸活组织检查和术后活组织取材检查甲状腺乳头状癌和非毒性结节性甲状腺肿COX-2基因表达无统计学差异(P>0.05)。结论COX-2对肿瘤转化具有重要作用。Objective To study the expression of COX-2 in papillary thyroid carcinoma (PTC), Hashimoto' s thyroiditis (HT), and nontoxic nodular goiter (NNG) and its effect. Methods Ninety patients with thyroid diseases including HT, FFC and NNG were collected. Total RNA was extracted from fine needle aspiration biopsy (FNAB) cytological material and postoperative tissues, cDNA synthesis in reverse transcription reaction and expression of COX-2 gene was analyzed by enzyme linked immunosorbent assay (ELISA). Results Expression lev- el of the COX-2 mRNA in PTC group was significantly higher compared with HT and NNG groups. Expression level of the COX-2 in PTC group was significantly higher than that in NNG group. There was no significant difference in COX-2 gene expression in the material obtained by FNAB and the postoperative thyroid tissue between PTC and NNG groups. Conclusion COX-2 gene plays an important role in neoplastic transformation.
