摘 要:为了了解油页岩矿区土壤环境中细菌群落的物种组成结构,以野外多点采集广东茂名油页岩矿区土壤样品作为研究对象,采用改进的SDS高盐提取法提取微生物总DNA,并利用PCR-DGGE技术分析细菌群落菌属种类。结果表明:DGGE指纹图谱中共包含13个操作分类单元(OTUs),它们涉及变形菌门(α-,β-,γ-)、厚壁菌门和绿湾菌门3个细菌门类,其中,α-变形菌有3个OTUs,分别属于Altererythrobacter,Chelativorans和Sphingomonas菌属;β-变形菌有3个OTUs,它们属于Hydrogenophaga和Thiobacillus菌属;γ-变形菌有4个OTUs,它们属于Acinetobacter和Pseudomonas菌属;厚壁菌门有2个OTUs,分别属于Faecalibacterium和Bacillus菌属;绿湾菌门有暂时无法确定菌属的1个OTU。Improved SDS-high-salt extraction method was used to extract microbial total DNA from the soil of multi-point collection. PCR-DGGE was applied to analyze the community structure of bacteria in soil from oil shale mine in Maoming of Guangdong Province. The results show that a total of thirteen operational taxonomic units(OTUs) are isolated from the PCR-DGGE patterns. Proteobacteria(α-, β-, γ-), Firmicutes and Chloroflexi are the dominant bacterial communities in this habitat. Ten OTUs which belong to seven genera of Proteobacteria including Altererythrobacter, Chelativorans, Sphingomonas, Hydrogenophaga, Thiobacillus, Acinetobacter and Pseudomonas are identified. Two OTUs which belong to two genera of Firmicutes including Faecalibacterium and Bacillus are identified, and one OTU which belongs to unidentified genus of Chloroflexi is also identified.
