摘  要:为明确小麦中F-box基因的存在种类及数量,作者通过数据库检索、编码区查找及功能结构域预测等途径,获得了NCBI和DFCI两大数据库中已报道的小麦F-box蛋白,并根据C端结构域特征对这些F-box蛋白进行了分类统计。数据库检索分类结果表明,在NCBI和DFCI数据库中共检索到符合条件的F-box基因38条,这些基因可以划分为F-box/LRR(17条,占总数的45%)、F-box/Kelch(6条,占总数的16%)和C端无明显结构域特征(15条,占总数的39%)3种类型。其中,F-box/LRR和C端无明显结构域类型所占比例较大。在此基础上,利用同源克隆策略在中国春小麦中获得了3条包含F-box结构域的序列,分别命名为Ta-SKP2A、Ta-FB和Ta-FBL14。Ta-SKP2A和Ta-FBL14属于F-box/LRR类型,Ta-FB仅包含一个F-box结构域,为C端无明显结构域类型。这些研究将为深入探讨F-box家族基因参与的小麦生物学过程及在其中的作用机理奠定基础。In order to identify the kind and number of F-box genes in wheat, F-box proteins were obtained from NCBI and DFCI databases according to the relative annotations. ORF a-nalysis and domain prediction were further carried out. Based on the different domains in the C-terminal of these wheat F-box homologues, we had classified these proteins into different types. Results from database annotations showed that 38 qualified F-box genes from NCBI and DFCI databases were obtained and classified into 3 types. The first group was F-box/LRR (17, accounting for 45%); the second was F-box/Kelch (6, accounting for 16%); the third was F-box protein without any obvious domains in the C-terminal (15, accounting for 39 ~). Majority genes of F-box family tested in the study belonged to F-box/LRR proteins and F-box protein without any obvious domains in C-terminal. Based on the former facts, 3 sequences with F-box domain in the N-terminal were obtained from the wheat cultivar Chinese Spring (CS) by homologous cloning strategy, and were designated as Ta-SKP2A, Ta-FB and Ta- FBL14. Ta-SKP2A and Ta-FBL14 belong to the F-box/LRR type, whereas Ta-FB is a F-box protein which contains no predicted domains in the C-terminal. These studies will facilitate further understanding of F-box family genes in biological process and their molecular mechanism.
