摘  要:目的:研究香椿子多酚提取物(polyphenols from toona sinensis seeds,PTSS)对神经毒素6-羟多巴胺(6-OHDA)诱导的PC12细胞损伤的保护作用。方法:体外培养PC12细胞,实验分为对照组、PTSS组、6-OHDA组及PTSS+6-OHDA组,倒置显微镜下观察各组细胞形态;MTT法检测细胞存活率;流式细胞术检测细胞凋亡情况;免疫组织化学染色法检测各组细胞Caspase-3、COX-2和TNF-α的表达变化。结果:PTSS可有效地降低6-OHDA引起的PC12细胞的损伤,增加细胞存活率,减少细胞早期凋亡;降低Caspase-3、COX-2及TNF-α的表达。结论:PTSS对6-OHDA引起的PC12细胞损伤具有明显的保护作用。Objective: To investigate the protective effects of PTSS on 6-OHDA induced injury in PC12 cells in vitro. Methods: PC12 cells were cultured. There were four groups: control group, PTSS group, 6-OHDA group and PTSS + 6-OHDA group. The change of cell morphology was observed under the inverted microscope. The cell viability was measured by MTT assay. The cell apoptosis was detected by flow cytometry. The expression of Caspase-3, COX-2 and TNF-α was detected using immunohistochemical method. Results: PTSS can effectively reduce 6-OHDA induced PC12 cell injury. Compared with the 6-OHDA group, the cell survival rate was increased, the cell apoptosis was reduced and the expression of Caspase-3, COX-2 and TNF-α was decreased in PTSS + 6-OHDA group. Conclusion: PTSS has a significant protective effect on 6-OHDA induced PC12 cell injury.
