摘 要:目的研究免疫抑制剂来氟米特对糖尿病肾病(DN)模型大鼠血清及肾组织白细胞介素-1β(IL-1β)、白细胞介素-6(IL-6)与肿瘤坏死因子-α(TNF-α)表达的影响。方法成年健康清洁级SD雄性大鼠45只,随机分为假手术组(15只)、模型组(15只)和来氟米特组(15只)。制作DN大鼠模型成功后,采用ELISA法检测血清IL-1β、IL-6、TNF-α水平,并采用RT-PCR方法检测3组大鼠肾组织IL-1β、IL-6、TNF-αmRNA表达。结果 ELISA法检测结果显示,模型组与来氟米特组大鼠血清IL-1β、IL-6、TNF-α均显著高于假手术组,差异有统计学意义(P<0.05);来氟米特组大鼠血清IL-1β、IL-6、TNF-α水平较模型组降低,差异有统计学意义(P<0.05)。RT-PCR检测结果表明,与假手术组大鼠比较,模型组和来氟米特组肾组织IL-1β、IL-6、TNF-αmRNA的表达量均显著升高,差异有统计学意义(P<0.05),而来氟米特组肾组织IL-1β、IL-6、TNF-αmRNA的表达则明显低于模型组,差异有统计学意义(P<0.05)。结论来氟米特可以抑制DN大鼠肾组织炎性因子IL-1β、IL-6、TNF-α表达,从而延缓DN进展。Objective To study the effect of Leflunomide on the expressions of interleukin-1β(IL-1β),interleukin-6(IL-6)and tumor necrosis factor-α(TNF-α)in serum and kidney of the diabetic nephopathy rat.Methods 45 healthy male rats were selected and randomly divided into the sham group(15),the model group(15)and treatment group.The diabetic nephropathy model was induced with Anderson′s method.The expressions of seurm IL-1β,IL-6and TNF-αwere detected by ELISA method,and the expressions of IL-1β,IL-6and TNF-αmRNA were observed by RT-PCR method.Results The serum levels of IL-1β,IL-6,TNF-αand mRNAs were increased significantly in model group and treatment group than that in sham group(P〈0.05),but decreased in treatment group than in model group(P〈0.05).Conclusion Leflunomide could down-regulate the expressions of IL-1β,IL-6and TNF-αin serum and kidney of the diabetic nephopathy rat and may delay the progress of DN.
