摘 要:目的:探讨植物甾醇对前列腺炎疼痛抑制作用的可能机制。方法:利用同源大鼠前列腺蛋白提取液辅以弗氏完全佐剂(FCA)诱导大鼠自身免疫性前列腺炎模型,植物甾醇灌胃给药10周后,测定大鼠水负荷0~4 h排尿量及尿离子浓度;ELISA法测定各组血清中IL-8、TNF-α及IL-10的含量变化;免疫组化法测定神经生长因子(NGF)在前列腺组织内原位表达的变化;Western Blot法测定前列腺组织内基质金属蛋白酶(MMP-9)及总p38丝裂原活化蛋白激酶(p38MAPK)表达的变化。结果:与假手术组比较,植物甾醇能不同程度减轻各组前列腺组织的炎症状况;植物甾醇能增加水负荷后2~4 h排尿量、促进同时段尿液中Na^+、Cl^-及Ca^(2+)的排出;模型组血清中IL-8、TNF-α、IL-10的含量明显增高,植物甾醇能明显降低大鼠血清中IL-8、TNF-α的水平;植物甾醇能明显抑制NGF、MMP-9及总p38MAPK在大鼠前列腺组织内的表达。结论:植物甾醇能很好地控制大鼠慢性非细菌性前列腺炎症,其可能的机制为通过抑制促炎因子及p38MAPK信号通路的某一环节,发挥其抗炎镇痛作用。AIM: To study the possible mechanisms of Phytosterols( PS) in neuralgia in chronic nonbacterial prostatitis( CNP) rats. METHODS:The CNP rat models were established by prostatic protein purification with Freund 'S completed adjuvant. We investigated diuresis effect by water-loaded rat test and measured the ion concentration in the urine. The changes of serum IL-8,TNF-α and IL-10 were evaluated by ELISA. The expression of NGF were evaluated by immunohistochemistry. The expression of MMP-9 and p38 MAPK were evaluated by western blot. RESULTS: PS could increase the concentration of Na+,Cl-and Ca2 +in the urine; PS could significantly raise urinary amount in 2-4 h,as compared with water-loaded model group; PS could obviously reduce the serumal content of IL-8,TNF-α; PS could obviously reduce prostate NGF,MMP-9and p38 MAPK expression. CONCLUSION: PS have good therapeutic effects on chronic nonbacterial prostatitis and could inhibit the release of inflammatory cytokines contribute to the maintenance of neuropathic pain,which maybe be concerned with that it could inhibit p38 MAPK signal pathway in prostategland of rats.
