摘 要:目的:研究减压内部沸腾法提取黄柏中盐酸小檗碱的最佳工艺参数。方法:以盐酸小檗碱得率为评价指标,采用单因素考察法,研究解吸液、提取压力、提取温度、提取时间等因素对提取效果的影响。并与乙醇回流提取法进行比较。结果:减压内部沸腾法提取黄柏中盐酸小檗碱的最佳工艺为:药材细粉加入55%的乙醇溶液60mL,解吸30min,加入60℃的水150mL,减压提取(真空度为0.06MPa,水浴温度60℃),提取时间10min,提取2次。结论:减压内部沸腾法提取黄柏中盐酸小檗碱提取效果优于乙醇回流提取法,具有快速、高效、低成本等优点。减压提取工艺稳定,可在生产中推广应用。Objective:To study the optimum process for extraction of berberine hydrochloride from phellodendron by decompressing inner ebullition. Methods: Taking berberine hydrochloride yield as measure index and using single factor investigation, the factors were studied, such as desorption liquid, extraction pressure, extraction temperature, extraction time, etc. The extraction results of bet berine hydrochloride from phellodendron by decompressing inner ebullition were compared with those by ethanol reflux extraction method. Results:The optimum extraction conditions were as follows: medicinal powder was added 55% ethanol solution 60mL, which the desorption time was 30 rain, then adding 150 mL 60℃ water,the extract under reduced pressure (vacuum degree of 0.06 MPa,water bath temperature 60℃), extraction time 10rain, extraction twice. Conclusion: The optimum process for extraction of berberine hydrochloride from phellodendron by decompressing inner ebullition was superior to the traditional extraction methods and had advantages of fast, efficient, low cost, etc. The optimum extraction process was stable, which could be applied in production.
