盐酸法舒地尔作用于增生性瘢痕的实验研究

摘　　要：目的：探讨盐酸法舒地尔对兔耳增生性瘢痕的抑制作用。方法随机将12只健康标准化日本大耳兔分为盐酸法舒地尔组（FH 组）、醋酸曲安奈德组（TAA 组）和生理盐水组（NS 组）共3组，每组4只，共24耳。将大耳兔麻醉后在兔耳建立增生性瘢痕模型，实验过程中对各组模型内分别注射等量实验药物。观察并记录给药过程中瘢痕的形状、大小、质地，取兔耳增生性瘢痕组织的标本进行 HE 染色，光镜下观察瘢痕组织成纤维细胞（Fi‐broblast ，FB）形态、分布以及数目，镜下通过测微尺计算各组标本中瘢痕增生指数（HI）。结果 FH 组及 TAA 组的兔耳增生性瘢痕较 NS 组明显变平、变软，颜色变淡，体积缩小。镜下可见 FH 组及 TAA 组瘢痕组织的真皮层有所增厚，但均较 NS 组薄，表皮细胞层数明显减少。 FH 组与 TAA 组的 HI 低于 NS 组，P ＜0．05，差异具有统计学意义。 FH 组和 TAA 组相比，P＞0．05，差异无统计学意义。结论盐酸法舒地尔注射液（钙离子拮抗剂）及醋酸曲安奈德溶液（肾上腺皮质激素）对兔耳增生性瘢痕均有明显抑制作用，本实验认为，盐酸法舒地尔注射液（FHI）作为新型钙离子拮抗剂，可作为临床辅助治疗增生性瘢痕的一类安全有效的治疗药物。Objective To study the effect of the fasudil hydrochloride injection on the scar models of the rab‐bits′ears .Methods 12 healthy standardization Janpanese rabbits ,a total of 24 ears are chosen to this experiment . They were divided into three groups which are FHI ,TAAI and NS groups .Each group were injected the same dose drugs during the experiment .Observeing and recording while in the process of drug scar shape ,size ,quality of a mate‐rial .After the rabbits were killed ,take hyperplastic scar tissue samples to HE staining .Caculate the number of the fi‐broblasts and the hypertrophic index through the medical microscope .The experimental datas and pictures will be statistic analysised by PASW Statistics 18 （SPSS 18 .0） statistical software .Results NS group still has the obvious skin contracture and the scars are more high than the others .The scars are dark qualitative ,hard ,subcutaneous of ob‐vious nodularity and can ,uneven surface .FH and TAA groups′ scar tissues slightly higher than the leather face ,did not see the obvious contracture ,.The color of the scars becomes shallow ,close to light ,quality of a material is become soft ,subcutaneous nodules small ,between the two groups scar atrophy no obvious difference degree .HE staining viewed by microscope ：there is a increased number of the capillaries and fibroblasts more than the FH and TAA groups .A loose orderly arrangement of collagen fibers ,unlike the collagen fibers arregment in the NS group .The nu‐cleus has not seen the obvious increase ,cells are arranged a close and orderly .The HI in the three groups ,we found that the HI of FH group is lower than it of the NS group ,the difference has a statistically significant （P〈 0 .05） .The HI of TAA group is lower than it of the NS group .Conclusion The fasudil hydrochloride injection（FHI） and the tri‐amcinolone acetonaide acetate injection（TAAI） have significant inhibitory effect on the hypertrophic scar models of＆amp;nbsp;the rabbits′ ears .They can make the s
