摘 要:目的:研究二冬膏对乌拉坦诱导肺肿瘤模型鼠TNF-α、IL-10、TGF-β1及肿瘤发生情况的影响。方法:小鼠随机分为6组,即正常组、模型组、二冬膏2.5g/kg组、二冬膏5g/kg组、二冬膏10g/kg组、六味地黄丸4.6g/kg组。除正常组外,其余各组采用小鼠腹腔注射乌拉坦800mg/kg,每周两次,连续五周,复制肺肿瘤小鼠模型。给药与造模同时进行,1次/d,连续15w。二冬膏10g/kg组、二冬膏5g/kg组、二冬膏2.5g/kg组、六味地黄丸4.6g/kg组分别给予等体积药物,正常组与模型组灌胃给予生理盐水。15w后,取血清和肺脏检测。解剖肺脏,观察肺肿瘤发生率及肿瘤结节数;双抗体夹心-酶联免疫吸附法检测血清TNF-α、IL-10和肺组织TGF-β1的含量;免疫组化检测肺组织TGF-β1活性的表达。结果:二冬膏能使模型鼠肺肿瘤发生率从86.7%降至40.0~66.7%,平均肿瘤结节数从7.32降至3.97~5.53,降低程度明显,并可明显降低血清中TNF-α、IL-10含量,使肺组织中TGF-β1含量和活性的表达也显著下降。结论:二冬膏能通过降低TNF-α、IL-10、TGF-β1的表达来延缓乌拉坦诱发肺肿瘤的发生发展。Objective:To study the effect of Erdong Gao (ED) on TNF-α, XL-10, TGF-β1 and tumorigenesis in mice with lung tumors by urethane. Methods:The clean KM mice were randomized i ntosix groups: the normal group(A) , the model group (B) , the Erdong Gao( 2.5 g/kg)group (C) ,the Erdong Gao(5g/kg)group(D), the Erdong Gao(10g/kg) group(E) and the Liuwei Dihuang Wan(4.6g/kg)group(F). In addition to A group mice, the others were induced lung tumors by urethane (0.8g/kg) subcutaneous injection,twice each week, which lasted for five weeks. Giving drug and establishing model at the same time, once a day, lasted for fifteen weeks. The C , D ,E ,F groups mice were respec- tively given ED (2.5 g/kg,5 g/kg,10 g/kg) and Liuwei Dihuang Wan (4.66 g/kg), the A , B groups were given normal saline. After fif- teen weeks, the mice were sacrificed, the serum and the lung tissue were detected. To observe the influence of ED on lung tumorigenesis and tumor nodules in mice by dissecting the lung. The contents of serum TNF-α,IL-10 and the contents of lung tissue TGF-β1 were detected by ELISA method. The expression of TGF-131 in lung tissue was determined by immunohistochemieal method. Results:In the process of lung car- cinogenesis,ED could significantly reduce lung chemical carcinogenesis ( dropped from 86.7% to 40. 0 - 66.7% ) and tumor nodules ( dropped from 7.32 to 3.97 ~ 5.53 ) in mice ( P 〈 0.05 or P 〈 0.01 ). ED could also significantly decrease the contents of serum TNF-α, XL-10 ( P 〈 0.05 or P 〈 0.01 ) and the expression of TGF-β1 in lung tissue ( P 〈 0.05 or P 〈 0. 01 ). Conclusions: ED could delay the occur- rence and development of lung carcinoma induced by urethane through decreasing the levels of TNF-α, XL-10, TGF-β1.
