摘 要:目的:研究乳香和没药药对的抑瘤作用,并考察其对肿瘤细胞中受体型酪氨酸激酶(RTKs)的影响,探讨乳没药对抑瘤作用的机理。方法:体外实验:采用乳没药对浸提液,MTT法分别测定其对2种人恶性肿瘤细胞株(SMMC7721、HL-60)增殖的影响,并检测其对肿瘤细胞RTKs活性的影响。体内实验:采用移植性荷瘤H22小鼠模型,接种3天后,荷瘤小鼠随机分组,并给予乳没药对10天,检测肿瘤重量并计算抑制率,同时检测肿瘤组织中RTKs的活性。结果:体外实验:乳没药对浸提液在5mg/ml^10mg/ml浓度下对两种肿瘤细胞的增殖均表现出明显的抑瘤作用,同时对2种肿瘤细胞的受体酪氨酸激酶活性有明显的抑制作用。体内实验:乳没药对在1.08g/kg和2.16g/kg剂量下对H22荷瘤小鼠肿瘤生长有明显的抑制作用,同时对肿瘤组织中受体酪氨酸激酶活性有明显的抑制作用。结论:乳没药对具有明显的抑瘤作用,其抑瘤机理可能与抑制肿瘤细胞内受体酪氨酸蛋白激酶途径有关。Objective: To study antitumous effects of receptor tyrosine kinases (RTKs) on SMMC7721 and HL-60 by Ru-Mo pair in vitro. Meth- od :Experment in vitro: Using leachate of Ru-Mo pair to treat diverse human malignant tumor cell strains (SMMC7721, HL-60)in vitro, pro- liferations of cells were detected by MTr assay, and the levels of RTKs by ELISA . Experment in vivo: The model of bearing H22 cancer mouse was established by transplantation, the mice were divided randomly into 5 groups : Ru-Mo high, medium and low dose group, cytoxan group and control group. After continuous treatment for 10 days, the tumors in each group were taken out and weighed for calculating inhibi- tion ratio. Meanwhile,the level of RTKs in tumors was detected by ELISA. Results: Experment in vitro: At the concentration of 5mg/ml and 10mg/ml, the leachate of Ru-Mo pair show obvious depressant effects of cell multiplication of human malignant tumor cell strains ( P 〈0.01, P 〈 0.05 ), and obvious depressant effects of RTKs on SMMC 7721 and HL-60 by Ru-Mo pair( P 〈 0.01 ; P 〈 0.05 ). Experment in vivo : At tile dosage of 1.08g/kg and 2.16g/kg, Ru-Mo pair shows obvious depressant effects of H22 mouse model ( P 〈 0.01, P 〈 0.05 ), and obvious depressant effects of RTKs on H22 by Ru-Mo pair( P 〈 0.01 ;P 〈 0.05 ). Conclusion: Ru-Mo pair shows obvious antitumous effects both in vitro and in vivo, the effect maybe correlate with depressing RTKs of malignant tumor cell.
