摘  要:目的:对芎归汤有效组分抗东莨菪碱致痴呆小鼠学习记忆能力减退的改善作用进行药效评价,并确定芎归汤有效组分对抗痴呆模型改善学习记忆药理学效应的"量-效"关系和ED50,为进一步开展药效评价提供剂量依据。方法:取昆明种小鼠进行Morris水迷宫、DT-200跳台筛选训练,将合格的小鼠80只,雌雄各半,按体重随机分为正常对照组、东莨菪碱致痴呆模型组、药效剂量1~5组和阳性药组,实验前分别给予各自治疗药物,连续给药30天后,除正常对照组外均腹腔注射东莨菪碱造模,30 min后进行Morris水迷宫实验及跳台实验,分析比较不同剂量组之间学习记忆能力差异性。结果:小鼠Morris水迷宫实验显示,芎归汤有效组分对东莨菪碱致痴呆小鼠学习记忆能力减退有明显改善作用,可以明显缩短小鼠水迷宫潜伏期,在10.92~43.68 g/kg剂量范围内均有显著性差异,且呈现一定的剂量依赖性,其ED50为4.79g/kg。小鼠跳台实验显示,芎归汤有效组分对东莨菪碱致痴呆小鼠学习记忆能力减退有明显改善作用,可以延长小鼠跳台潜伏期和减少跳台错误次数,在2.73~43.68 g/kg剂量范围内均有显著性差异,且呈现一定的剂量依赖性,其ED50为2.70g/kg。结论:芎归汤有效组分对东莨菪碱致小鼠学习记忆能力减退有明显的改善作用,并且随着剂量的增加药效越来越明显,呈现一定的剂量依赖性,具有明显的"量-效"关系。Objective: By studying the effect of Xiong Gut Tang in treating hypomnesis in Morris water maze and step down test in Dementia Model induced by scopolamine, in order to determine the dose-effect relationship and ED50, and offer the references and research ideas for the fur- ther experiments, Methods : The normal mice were screening and training with Morris water maze and step down test before the formal experi- ment. And the qualified mice were divided into different groups such as the normal group, the model group, the different dose group and sposi- tive drug group. The ability of learning and memory were observed though using Morris water maze and step down test after 30 days adminis- trated. Results : By analyzing the results of each group of mice in Morris water maze, the effective components from Xiong Gut Tang had a protective effect on the treatment of hypomnesis. Compared with the model group, the Xiong Gut Tang has a significant differences in the dose range of 10.92 - 43.68g / kg, and the ED50 is 4. 7907g/kg. By analyzing the results of each group of mice in step down test, the effective components from Xiong Gut Tang had a protective effect on the treatment of hypomnesis. Compared with the model group, the Xiong Gut Tang has a significant differences in the dose range of 2.73 ~ 43.68g/kg, and the ED50 is 2. 7046g/kg. Conclusion: The effective components from Xiong Gut Tang had a protective effect on the abihty of learning and memory in dementia model induced by scopolamine and this show an obvious dose-effect relationshio.
