摘  要:目的:评估脑组织线粒体呼吸功能作为轻度认知障碍(mild cognitive impairment,MCI)药效学评价指标的可能性。方法:采用自然衰老(16月龄)大鼠和东莨菪碱所致记忆障碍大鼠进行morris水迷宫实验,以评估其宏观空间学习记忆损伤情况;检测两种模型动物脑组织SOD活性和MDA含量,并利用Clark氧电极法对其脑组织线粒体呼吸功能进行评价。结果:与正常成年动物相比,两种记忆障碍模型动物在水迷宫实验中表现为逃避潜伏期延长,穿越平台次数降低,以及在原平台所在象限游泳时间比例减少,脑组织SOD活性降低、MDA含量增高,线粒体Ⅲ态呼吸速率、RCR及P/O、OPR值均明显下降。结论:线粒体呼吸功能损伤可在早期自然衰老和东莨菪碱致记忆障碍大鼠模型脑组织中被检测,是一种较灵敏的检测指标,并可与脑组织过氧化和宏观空间学习记忆能力损伤等指标共同用于MCI的药效学研究。Objective: To evaluate the possibility of pharmaeodynamic evaluation index of mitochondrial respiratory function in brain tissue as mild cognitive impairment (MCI). Methods: Using Morris water maze test to evaluate the macro spatial learning and memory impairment in Natu-ral aging animal models and scopolamine animal models, and survey the contents of SOD and MDA of the brain. Evaluation of mitochondria respiratory function of brain in animals with cognitive impairment by Clark oxygen electrode method. Results: Compared with the normal ani- mal, Natural aging animal models and scopolamine animal models of cognitive impairment have longer incubation period and shorter time into the platform,proportion of former platform quadrant swimming time is also significantly reduce. The content of SOD was significantly de- creased and MDA was increased in brain. The R3, RCR, P/O and OPR of brain mitochondria are significantly decreased. Conclusion: Mi- tochondrial respiratory function damage is a more sensitive test indicators, and it can be detected in natural aging animal models and scopol- amine animal models. The damage of mitochondrial function can be used to study the effects of MCI on the effects of brain tissue peroxidation and macro spatial learning and memory deficits.
