摘  要:目的探讨血清可溶性CD 3 0(s CD 3 0)作为电离辐射剂量估算的生物指标的可行性。方法健康雄性C57BL/6小鼠300只,随机分为对照组及1、3、5、7Gy辐照组(n=60)。辐照组小鼠接受60Coγ射线一次性全身辐照,剂量率为0.78Gy/min。各组分别于辐照后24、48、72h留取血清样本(每组每时间点20只小鼠),采用定量抗体芯片检测s CD30浓度,并通过回归分析法分析血清s CD30浓度与辐照剂量的相关性。结果未辐照小鼠s CD30正常值范围为121.2±25.6pg/ml;辐照后24、48、72h,血清s CD30浓度随辐射剂量增大而降低。回归分析显示,血清s CD30浓度与辐照剂量具有明显的剂量-效应关系(R^2=0.9913),其回归方程为y=1.1489x^2-21.139x+121.44。结论小鼠血清s CD30表达基线稳定,其浓度变化与急性辐照剂量相关,可作为γ电离辐射剂量估算的生物指标。Objective To assess the feasibility of serum soluble CD30(s CD30) as a biomarker for radiation exposure. Methods Three hundred male C57BL/6 mice were prepared and randomly divided into 4 radiation groups(60 each) and control(n=60). Mice in radiation groups were treated with 60 Co total-body irradiation in the dose of 1, 3, 5 or 7 Gy, and the dose rate of 60 Co γ ray at one-time whole-body irradiation was 0.78 Gy/min. 24, 48, or 72 h after radiation, the serum samples were obtained(20 mice in each group at each time point). The serum s CD30 concentrations were analyzed with custom-made antibody array, which was purchased from Ray Biotech. To further analyze the relationships of serum s CD30 concentration and radiation dose, regression analysis was used. Results The baseline level of s CD30 in serum of non-exposed animals was 121.2±25.61pg/ml. Moreover, serum s CD30 concentration was inversely related to the radiation dose during 72 h after exposure. Regression analysis showed an obvious dose-effect relationship between serum s CD30 concentration and radiation dose(24h after radiation, R^2=0.8715; 48 h after radiation, R^2=0.9024; 72 h after radiation, R^2=0.9913). Using these equations, radiation exposure can be easily estimated by the serum s CD30 value. Conclusions The baseline level of serum s CD30 remains steady without radiation. Further regression analysis uncovers a strong correlation between serum s CD30 concentration and radiation dose, and serum s CD30 may be used to estimate radiation exposure as a biomarker.
