摘 要:为确定中药与环丙沙星联用对鸡源性沙门菌的抑菌作用,采用微量稀释法测定黄连、黄芩、五倍子、丁香醇提取物的最低抑菌浓度(MIC),筛选出抑菌作用较强的中药提取物,并用微量棋盘稀释法测定其与环丙沙星联用对沙门菌的部分抑菌浓度(FIC)。结果显示,4味中药醇提物对沙门菌都有一定的抑菌作用,黄连相对于其他3味中药具有较好的抑菌效果,对黄连醇提物按照黄连素提取工艺进一步提取,获得粗品,此次提取物提取率为8.17%。联合药敏试验结果显示,黄连提取物在1.28μg/mL时,使环丙沙星的MIC值降低至0.64μg/mL;而环丙沙星在1 562μg/mL时,黄连提取物的MIC值降低至781μg/mL,说明两药联合应用可明显增强环丙沙星或黄连的抑菌效果,而根据FIC指数判断标准可得FIC指数为0.75,这表明黄连提取物与环丙沙星体外联合抑菌作用表现为相加作用。To observe the antibacterial effect of Chinese herbal medicine combined with ciprofloxacin (CIP) on Salmonella isolated from chickens, minimum inhibitory concentration (MIC) of alcohol extract of goldthread, scutellaria, gallnut, clove against Salmonella were determined by micro dilution method. One extract with strong antibacterial effect was selected and fractional inhibitory concentrations (FIC) of the extract combined with CIP were determined by constantcheckerboard dilution method. The resuhs showed that all 4 extracts had antibacterial effect against Salmonella. Compared with other 3 kinds of extracts, goldthread extract had a better effect. Further extraction was done according to the extraction process of berberine,crude product was obtained and the extraction rate was 8.17%. The test of FIC showed that, when the extract concentration was at 1.28 μg/mL, the MIC of CIP was reduced to 0.64 μg/mL; and when CIP was at 1 562 μg/mL concentration, the MIC of the extract was reduced to 781 μg/mL. Thus, the combination could significantly enhance the antibacterial effect of ciprofloxacin or goldthread. According to the judgment standard, the FIC index was 0.75. It indicated that the antibacterial activity of goldthread extract combined with CIP in vitro pefformanced as additive effect against Salmonella.
