摘 要:目的研究白术七物颗粒对气阴两虚型结肠慢传输型便秘(STC)大鼠结肠组织血管活性肠肽(VIP)及P物质(SP)的影响,初步探讨白术七物颗粒对气阴两虚型STC的治疗作用及其作用机制。方法 2014年10—12月,采用随机数字表法将60只SD大鼠分为正常组、模型组、治疗组,各20只。模型组、治疗组予以皮下注射盐酸吗啡注射液,同时灌服青皮、枳壳、附子配方颗粒,建立气阴两虚型STC模型;正常组皮下注射等量等渗0.9%氯化钠注射液。记录造模期间大鼠的一般状况,造模前、后24 h各组大鼠粪便干重,肠道炭末推进率。造模成功后,治疗组大鼠灌服白术七物颗粒水溶液,其余两组灌服等量等渗0.9%氯化钠溶液。给药14 d后检测各组大鼠结肠组织VIP、SP水平。结果正常组大鼠运动灵活,饮食、饮水、大小便正常;其余两组大鼠造模后出现懒动、毛色枯黄、大便次数减少、颗粒变大且质硬等情况。造模前3组大鼠粪便干重比较,差异无统计学意义(P>0.05)。造模后模型组、治疗组大鼠粪便干重低于正常组(P<0.05)。模型组、治疗组大鼠肠道炭末推进率小于正常组(P<0.05)。模型组、治疗组大鼠VIP水平高于正常组(P<0.05);治疗组大鼠VIP水平低于模型组(P<0.05)。模型组、治疗组大鼠SP水平低于正常组(P<0.05);治疗组SP水平高于模型组(P<0.05)。结论白术七物颗粒能有效治疗气阴两虚型STC,其作用可能与下调VIP水平、升高SP水平有关。Objective To investigate the influence of Baizhu Qiwu granules on vasoactive intestinal peptide (VIP) and substance P (SP) of colon tissue of rats with colonic slow transit constipation (STC) of qi and yin deficiency, and to preliminarily investigate the efficacy and mechanism of Baizhu Qiwu granules in the treatment of STC of qi and yin deficiency. Methods From October to December in 2014, 60 SD rats were divided into normal group ( n = 20) , model group ( n = 20) and treatment group (n = 20) by random number table method. Model group and treatment group were administrated with morphine hydrochloride injection by subcutaneous injection and were also administrated with formula granules composed of pericarpium citri reticulatae viride, fructus aurantii and aconitum carmichaelii to build STC models of qi and yin deficiency; normal group was administrated with the same amount of 0. 9% sodium chloride injection by isotonic subcutaneous injection. The general condition of the rats was recorded; before model building and 24 hours after model building, the condition of dry weight faeces and the rate of charcoal propulsion in intestinal tract were observed. After successful model building, the treatment group was administrated with water solution of Baizhu Qiwu granules by drench. The other two groups were administrated with the same amount of 0. 9% sodium chloride injection by isotonic subcutaneous injection. The VIP and SP levels in colon tissues of the rats were detected 14 days after drug administration. Results The rats of the normal group had flexible motion and normal diet, drinking and defecation; the rats of the other two groups behaved sluggish in moving, with their hair dry and yellow, the amount of stool decreasing and the stool particle being smaller and harder. Before model building, the three groups were not significantly different in the number of rats having dry weight feces ( P 〉 0. 05 ) . After model building, the model group and the treatment group were lower than normal
