摘 要:目的探讨大青龙汤对流感病毒感染小鼠血清及肺组织中免疫因子的影响及免疫学机制。方法选取5~8周龄Babl/c小鼠48只,SPF级,雌雄各半,体质量18~23g;采用流感病毒亚甲型鼠肺适应株(FM/1/47株)建立小鼠感染模型,将小鼠正常饲养7d后随机分为正常组、模型组、大青龙汤低剂量组和高剂量组,每组12只;各组小鼠均采用灌胃给药,每日1次;大青龙汤低剂量为9倍处方量,高剂量为27倍处方量,正常对照组和模型对照组给等容积生理盐水,给药时间从建模后第2天开始连续给药5d,停药后第2天留取标本,测定各组小鼠血T淋巴细胞亚群、血清及肺组织中TNF-α、IFN-γ、IL-6、IL-10水平变化。结果各组小鼠T淋巴细胞亚群检测CD4+和CD4+/CD8+在各组间差异有统计学意义(P〈0.05),大青龙汤高剂量对小鼠血中CD4+和CD4+/CD8+降低有一定的抑制作用,差异有统计学意义(P〈0.05);大青龙汤低剂量和高剂量组可明显升高小鼠血清中TNF-α,IFN-γ水平及IL-10水平,降低IL-6水平;大青龙汤低剂量和高剂量组可明显降低小鼠肺组织中TNF-α和IFN-γ水平,对IL-6和IL-10水平升降作用不明显。结论大青龙汤对流感病毒感染引起的细胞免疫功能降低有一定的抑制作用,可以增强全身免疫,并抑制病变肺脏的过度免疫。OBJECTIVE To explore the effect of Daqinglong soup on immune factors in serum and lung tissues in mice with influenza virus infection and to explore the immunological mechanism.METHODS Totally 48SPF-grade Babl/c mice(half males and half females)were selected,aged 5-8weeks and weighted 18-23 g.The mouse lung adapted strain(FM/1/47strain)of influenza virus was used to establish a murine model of infections.After 7days of normal feeding,the mice were randomly divided into the normal group,the model group,the low dose group and the high dose group,12 mice in each group.Each group received gastric administration once a day.The low dose was 8times higher than the prescription dose of Daqinglong soup and the high dose was 26 times higher than the prescription dose.The normal control group and the model control group received normal saline of equal volume.The five-day continuous administrated began at the second day after modeling and mice were sample at 2days after drug withdrawal to determine the changes in TNF-α,IFN-γ,IL-6and IL-10 levels in blood T lymphocyte subsets,serum and lung tissues.RESULTS There was significance in CD4+and CD4+/CD8+in T lymphocyte subsets between groups(P〈0.05),indicating Daqinglong soup had certain inhibition effect on CD4+and CD4+/CD8+in mouse blood(P〈0.05).TNF-α,IFN-γand IL-10 levels were significantly increased and IL-6was decreased in serum in both low and high dose groups.TNF-αand IFN-γlevels were significantly decreased in lung tissues in these two groups,however,the effect on IL-6and IL-10 levels in lung tissues was not obvious.CONCLUSION Daqinglong soup has certain inhibitory effect on decreased immune function of cells caused by influenza virus infections.It can enhance the body immunity and inhibit excessive immunity in lungs with lesions.
