摘 要:目的探讨β_1受体阻滞剂对力竭运动大鼠心肌能量代谢变化的影响及其对应激心肌的保护机制。方法雄性Wistar大鼠36只,随机分为对照组、一次力竭组、一次力竭+倍他乐克治疗组,每组各12只。游泳力竭后测定心肌ATP、血浆肾上腺素(epinephrine,E)、去甲肾上腺素(norepinephrine,NE)、游离脂肪酸(free fatty acid,FFA)浓度,荧光定量逆转录聚合酶链反应(RT-PCR)及Western blot法分析心肌线粒体解偶联蛋白2(uncoupling protein 2,UCP2)表达。结果与对照组相比,一次力竭组、一次力竭+倍他乐克治疗组血浆FFA、E、NE浓度均增高,心肌ATP含量均降低(P<0.05)。RT-PCR结果表明一次力竭组、一次力竭+倍他乐克治疗组UCP2m RNA表达量较对照组分别增加64%、43%(P均<0.05)。Western blot结果表明一次力竭组、一次力竭+倍他乐克治疗组UCP2表达量较对照组分别增加78%、42%(P均<0.05)。相关分析显示血浆FFA浓度与血浆E、NE水平呈显著正相关(r=0.93,r=0.88,P<0.01),心肌组织UCP2表达量与血浆FFA浓度呈显著正相关(r=0.98,P<0.01)。结论应用β1受体阻滞剂后可降低运动应激对心肌能量代谢的不利影响,减轻心脏负荷。Objective To discuss the influence of β1 receptor blocker on changes of myocardial energy metabolism and protective mechanism to stress myocardium in rats after exhaustive exercise. Methods Male Wistar rats(n=36) were randomly divided into control group, once exhaustion group and once exhaustion+metoprolol group(treatment group, each n=12). The concentrations of myocardial ATP, plasma epinephrine(E), norepinephrine(NE) and free fatty acid(FFA) were detected after exhaustive swimming. The expression of myocardial mitochondrial uncoupling protein 2(UCP2) was analyzed by using RT-PCR and Western blot.Results Compared with control group, the concentrations of plasma FFA, E and NE increased and myocardial ATP decreased in once exhaustion group and treatment group (P〈0.05). The results of RT-PCR showed that the expression of UCP2 mRNA increased by 64% in once exhaustion group and by 43% in treatment group compared with control group (all P〈0.05). The results of Western blot showed that UCP2 expression increased by 78% in once exhaustion group and by 42% in treatment group compared'with control group (all P〈O.05). Correlation analysis showed that concentration of plasma FFA was positively correlated to levels of plasma E and NE (r=0.93, r=0.88, P〈O.01), and UCP2 expression was greatly positively correlated to concentration of plasma FFA (r=0.98, P〈0.01). Conclusion The application of β 1 receptor blocker can reduce adverse impact of exercise stress on myocardial energy metabolism, and relieve cardiac overload.
