摘 要:目的观察益气化瘀方对子宫复旧不全大鼠子宫内膜细胞凋亡及Bcl-2、Bax表达的影响,探讨其作用机制。方法 40只分娩后SD大鼠随机分为空白组、模型组、新生化组和益气低、高剂量组。除空白组外,分娩后,将过夜培养的大肠杆菌进行子宫内接种制备大鼠炎症产后子宫复旧不全模型,并于造模后第1-7日分别给予相应药物灌胃。HE染色观察各组大鼠子宫切片病理变化;免疫组化检测子宫内膜组织Bcl-2、Bax蛋白表达。结果模型组Bcl-2蛋白表达明显低于空白组(P<0.01),Bax蛋白表达明显高于空白组(P<0.01);益气低、高剂量组和新生化组Bcl-2蛋白表达较模型组均明显升高(P<0.01),Bax蛋白表达均明显降低(P<0.01);益气低剂量组Bcl-2蛋白表达较新生化组明显降低(P<0.01),Bax蛋白表达较新生化组明显升高(P<0.01)。结论益气化瘀方可能通过上调Bcl-2、下调Bax蛋白表达,降低子宫内膜细胞凋亡的发生,促进子宫内膜增生修复,从而促进产后子宫复旧。Objective To investigate the effects of Yiqi Huayu Decoction on cell apoptosis and Bcl-2, Bax expressions of incomplete puerperal involution of uterus; To discuss its mechanism of action. Methods Forty SD rats were randomly divided into normal group, model group, Xinshenghua group, high- and lower-dose Yiqi groups. The first day after delivery, except for the normal group, rats received intrauterine inoculation with overnight cultured diluted E. coli to establish rat models with incomplete puerperal involution of uterus. They were given corresponding medicine for gavage during day 1-7 after the modeling. Pathological changes in rat uterus slice were observed by HE staining in each group; immunohistochemical detection was used to detect protein expressions of Bcl-2 and Bax in endometrial tissue. Results The protein expression of Bcl-2 in the model group was lower than that in the normal group(P<0.01), and protein expression of Bax in the model group was higher than that in the normal group(P<0.01). The protein expression of Bcl-2 in the high- and lower-dose Yiqi groups and Xinshenghua group was higher than that in the model group(P<0.01), and protein expression of Bax in the high- and lower-dose Yiqi groups and Xinshenghua group was lower than that in the model group(P<0.01). The protein expression of Bcl-2 in the lower-dose Yiqi group was lower than that in Xinshenghua group(P<0.01), and protein expression of Bax in the lower-dose Yiqi group was higher than that in Xinshenghua group(P<0.01). Conclusion Yiqi Huayu Decoction can promote the repair of endometrial hyperplasia and postpartum uterine involution through up-regulating Bcl-2 protein expression and down-regulating Bax protein expression, and reducing the incidence of endometrial cell apoptosis.
