芪贞固本方对Lewis肺癌小鼠转移的抑制作用及机制研究

摘　　要：目的:研究芪贞固本方对Lewis肺癌小鼠的转移抑制作用及作用机制。方法:建立Lewis肺癌小鼠模型,按实验分组干预,计算比较各组抑瘤率、肺转移抑制率,免疫组化法检测E-钙黏素表达,RT-PCR法检测VEGFmRNA表达。结果:芪贞固本方有一定的抑瘤及肺转移抑制作用,抑瘤率及肺转移抑制率分别为:13.97%、19.13%;与对照组相比,芪贞固本方可明显下调VEGFmRNA的表达,促进E-钙黏素的表达(P<0.05),与顺铂合用时作用增强。结论:芪贞固本方可抑制Lewis肺癌小鼠转移,其机制可能是促进E-钙黏素的表达,下调VEGFmRNA的表达。Objective： To study the anti- metastasis effect and mechanism of Qizhen Guben Formula on mice model with Lewis lung cancer. Methods： To establish the Lewis lung cancer model in mice and interven the mice according to their groups. To calculate and compare the rate of anti- tumor and anti- metastasis. The expression of E- cadherin in tumor cells was measured by immunohistochemistry and the expression of VEGFmRNA detected by real- time PCR. Results： Qizhen Guben Formula had a certain extent effect of anti- tumor and anti- metastasis. The rates of anti- tumor and anti- metastasis were 13. 97% and 19. 13% respectively. Compared to the control group,Qizhen Guben Formula can down- regulate the expression of VEGFmRNA and promote the expression of E- cadherin significantly（ P〈0. 05）. And the effect would increase together with DDP. Conclusion： Qizhen Guben Formula can inhibite the metastasis of mice with Lewis lung cancer and the mechanism may be related to promoting the expression of E- cadherin and down- regulating the expression of VEGFmRNA.
