摘 要:目的研究柚皮苷对蛛网膜下腔出血(SAH)引起的脑血管痉挛以及神经元凋亡的影响,并分析其潜在机制。方法 60只SD大鼠随机分为假手术组(Sham组)、SAH模型组(SAH组)、柚皮苷治疗组(Nar组,80 mg/kg),每组20只。各组10只大鼠取患侧大脑皮质匀浆液,检测MDA含量;另10只大鼠断头取脑,固定后常规包埋后切片,HE染色测基底动脉内径周长。TUNEL和NEUN免疫荧光双染检测大鼠脑皮质神经元凋亡。结果与模型组比较,柚皮苷能明显降低SAH氧化应激指标MDA的水平(P<0.01);并显著抑制脑基底动脉血管痉挛(P<0.01);免疫荧光结果显示,柚皮苷能显著减少SAH大脑皮质神经元凋亡的数量(P<0.01)。结论柚皮苷可以减轻大鼠SAH后早期脑血管痉挛,减少大脑皮质神经元凋亡的功能,其机制可能与抑制SAH后氧化应激有关。Objective To study the effect of naringin on cerebral vasospasm and neuronal apoptosis induced by subarachnoid hemorrhage in rats,and analyze the potential mechanisms.Methods Sixty SD rats were randomly divided into sham operation group(Sham group),SAH group,and naringin group(Nar group,80 mg/kg),20 rats in each group.Among 10 rats in each group,ipsilateral hemispheres of brains were homogenized to detect the MDA level.Another 10 rats' brains were dissected and fixed-embedded sections,HE staining was conducted to detect the basilar artery diameter.The neuronal apoptosis was detected by immunofluorescent double staining with TUNEL and NEUN antibody.Results Compared with SAH group,naringin significantly attenuated the level of MDA-an oxidative stress marker(P〈0.01),and inhibited cerebral vasospasm(P〈0.01).The result of immunofluorescent double staining showed that naringin significantly attenuated the neuron apoptosis in brain cortex(P〈0.01).Conclusion Naringin can attenuate cerebral vasospasm and decrease the number of neuron apoptosis in brain cortex in rats after SAH,the potential mechanism may be related with anti-oxidant after SAH.
