正常肝脏与乙型肝炎肝硬化及乙型肝炎病毒相关性肝癌组织中miRNA表达谱差异

摘　　要：目的探讨正常肝脏、乙型肝炎肝硬化、乙型肝炎病毒(HBV)相关性肝癌组织中miRNA表达谱差异,以期将有效依据提供给了临床对HBV感染发展为肝硬化,进而发展为肝癌这一进程中miRNA的表达变化的进一步阐明。方法回顾性分析2013年1月-2014年9月医院收治的接受肝癌切除及肝组织活检的患者临床资料,运用miRNA芯片对正常肝脏、乙型肝炎肝硬化、HBV相关性肝癌组织中的miRNA表达进行检测。结果正常肝脏、乙型肝炎肝硬化、HBV相关性肝癌组织标本总RNA的A260/A280值分别为1.88、1.85、1.83,均具有较为清晰的18S和28S条带,密度之比约为1∶2,质量较好,后续的miRNA芯片检测及PCR实时定量的试验要求能够得到有效满足;乙型肝炎肝硬化、HBV相关性肝癌组织中表达比正常肝脏组织上调和下调至少两倍的miRNA分别有6个和8个,实时定量PCR结果和miRNA芯片结果基本相同,miRNA芯片结果可信。结论正常肝脏、乙型肝炎肝硬化、HBV相关性肝癌组织中miRNA表达谱差异主要体现在进程早期。OBJECTIVE To investigate the differences of miRNA expression profiling in normal liver,liver cirrhosis,Hepatitis B virus（HBV）-related liver cancer tissues,so as to provide an effective basis for the further elucidation of miRNA in the process of clinical HBV infection developed into cirrhosis,liver cancer and further development of the expression.METHODS The clinical data of liver cancer and liver biopsy patients in our hospital from Jan.2013 to Sep.2014 were statistically analyzed,the normal liver cirrhosis,HBV-related liver cancer tissue miRNA expression were detected by miRNA microarray.RESULTS A260/A280 values of total RNA of normal liver,liver cirrhosis,HBV-related liver cancer tissue samples were 1.88,1.85,1.83,all with relatively clear 18Sand28 Sbands,and the density ratio was about 1：2with good quality.Subsequent miRNA microarray and quantitative real-time PCR experiments requires were effectively met;cirrhosis,HBV-related liver cancer tissues regulated up and down at least 2-fold miRNA than normal liver tissue were 6and 8,and real-time quantitative PCR results and miRNA microarray results were basically the same. The miRNA microarray results were credible.CONCLUSION.The differences of miRNA expression profiling in normal liver,liver cirrhosis,HBV-related liver cancer tissues are mainly reflected in the early process.
