摘 要:【目的】探讨植物雌激素类药物金雀异黄素(genistein,GEN)对骨肉瘤细胞U2OS生长的影响作用。【方法】用不同浓度的GEN处理U2OS细胞,分别在24、48和72 h采用MTT法检测细胞增殖抑制情况,用酶活性法检测碱性磷酸酶(alkaline phosphate enzyme,AKP)活性和ELISA法检测骨钙素(osteocalcin,BGP)的表达。【结果】MTT检测显示,GEN对U2OS细胞生长的影响与GEN作用时间及浓度有密切关系,当GEN浓度增加至10 000 nmol/L时,OD值远低于对照组(P<0.05),而在低浓度(0.1 nmol/L和1 nmol/L)GEN作用72 h后,OD值又远高于对照组(P<0.05);AKP活性检测表明,高浓度(10 000 nmol/L)GEN作用48 h和72 h能明显抑制AKP的活性(P<0.05),而低浓度(0.1 nmol/L)GEN却能增加AKP的活性(P<0.05);ELISA实验表明,GEN处理U2OS细胞后对其BGP表达的影响作用也同样表现为与药物浓度及培养时间有关,作用48、72 h后,BGP的表达随着药物浓度的降低却有明显的增加。【结论】GEN除了能抑制U2OS细胞的增殖、具有抗肿瘤作用外,低剂量应用时具有促进成骨分化作用,其作用机制可能与其激活雌激素受体有关。【Objective】To explore the effect of genistein(GEN), one of plant phytoestrogens, on osteosarcoma cells U2 OS.【Methods】U2OS cells were cultivated with GEN in different concentrations. After 24 h, 48 h and 72 h respectively, the status of cell proliferation was detected by MTT assay; alkaline phosphatase(AKP) activity was detected; the expression of osteocalcin(BGP) was detected by ELISA.【Results】MTT assay showed that the effect of GEN on the growth of U2 OS cells was closely related to the duration and concentration of GEN; when the concentration of GEN was increased to 10 000 nmol/L, OD values were much lower than those in the control group(P0.05); when GEN was at low concentrations(0.1 nmol/L and 1 nmol/L) for 72 h, its OD values were far higher than those in the control group(P0.05). AKP activity assay showed that GEN at high concentration(10 000 nmol/L) for 48 h and 72 h can inhibit the AKP activity obviously(P0.05), whereas AKP activity was markedly improved when GEN was in low concentration(0.1~100 nmol/L)(P0.05). ELISA experiments indicated that the expression of BGP in U2 OS cells treated with GEN was also relevant to the drug concentration and duration; and there was a significant increase in the expression of BGP when the drug concentration decreased after 48 h and72 h.【Conclusion】GEN can not only inhibit the proliferation of U2 OS cells but also have an anti-tumor effect, however GEN in a low dose can promoted osteogenic differentiation. Its mechanism may be relevant to activating estrogen receptor(ER).
