摘 要:目的分析子痫前期孕妇胎盘剥离面母体血液中自噬蛋白Beclin-1、LC3-II的表达量及胎盘CD8^+T细胞浸润程度,并探究两者间的相关性。方法 30例正常孕妇作为对照组,28例重度子痫前期孕妇与34例轻度子痫前期孕妇作为实验组。应用ELISA法检测各组孕妇胎盘剥离面母体血液中自噬蛋白Beclin-1、LC3-II的表达量,流式细胞术分析胎盘CD8^+T细胞数量,并对两组数据进行相关性分析。结果实验组孕妇血清自噬蛋白Beclin-1、LC3-II的表达量及胎盘CD8^+T细胞数量均明显高于对照组(P<0.01),其中重度子痫前期组各检测结果均显著高于轻度子痫前期组(P<0.01);实验组Beclin-1蛋白表达量与CD8^+T细胞数呈正相关,其中轻度子痫前期组两者间相关系数r=0.495,P<0.01,重度子痫前期组两者间相关系数r=0.563,P<0.01;实验组LC3-II蛋白表达量与CD8^+T细胞数呈正相关,其中轻度子痫前期组两者间相关系数r=0.637,P<001,重度子痫前期组两者间相关系数r=0.584,P<0.01。结论子痫前期孕妇血清自噬蛋白Beclin-1、LC3-II表达量及胎盘CD8^+T细胞数量均高于正常孕妇,相关数值随着发病程度加重而升高,自噬蛋白水平及胎盘CD8^+T细胞数量呈正相关。Objective To analyze the expression of autophagic proteins Beclin-1 and LC3-II in placenta peeling surface maternal blood of preeclampsia and placental CD8^+ cell infiltration,and explore the correlation between autophagic protein expression and CD8^+ cell infiltration. Methods 30 cases of normal pregnant women were collected as the control group,28 cases of severe preeclampsia and 34 cases of pregnant women with mild preeclampsia were collected as the experimental groups. ELISA was used to detect the levels of autophagic proteins Beclin-1 and LC3-II. Flow cytometry was used to analyze the number of CD8^+ cells in the placenta. The correlation between autophagic protein expression and CD8^+ cell infiltration was analyzed. Results Compared with the control group,the expression of autophagic proteins Beclin-1 and LC3-II and placenta CD8^+ cell infiltration were significantly increased in both experimental groups( P〈0.01). Notably,these data were higher in the severe preeclampsia group than that of the mild preeclampsia group( P〈0. 01). Beclin-1 protein expression was positively correlated with placental CD8^+ cell infiltration in both experimental groups. The correlation coefficient in mild preeclampsia group was 0.495( P〈0.01),and in severe preeclampsia group was 0.563( P〈0.01). LC3-II protein expression was positively correlated with placental CD8^+ cell infiltration in both experimental groups. The correlation coefficient in mild preeclampsia group was 0. 637( P〈0. 01),and in severe preeclampsia group was 0.584( P〈0.01). Conclusion The expression of autophagic proteins Beclin-1 and LC3-II and placenta CD8^+ cell infiltration in preeclampsia are higher than that of the normal pregnant women,which will increase as the disease progress. There is a positive correlation between autophagic protein expression and CD8^+ cell infiltration.
