超微风眩宁方治疗高血压病肝阳上亢证的实验研究

摘　　要：目的：观察风眩宁治疗高血压病的效果,同时对比超微颗粒与传统汤剂的疗效,探讨该方的作用机制。方法：采用左肾动脉狭窄手术制备高血压病肝阳上亢证大鼠模型,按血压分层将造模成功的大鼠随机分为6组：模型组,氯沙坦钾胶囊组,天麻钩藤颗粒组,风眩宁方超微高、低剂量组和风眩宁方传统剂量组,每组10只,另取12只为假手术组。各组分别给予相应药物进行干预,假手术组及模型组均给予同容积蒸馏水灌胃。观察比较各组大鼠血压的变化情况;用酶学方法测定大鼠空腹血糖（FPG）、放射免疫分析法测定空腹胰岛素（FINS）水平、计算胰岛素敏感指数（ISI）;取出大鼠双侧肾脏组织,进行显微形态学观察。结果：造模后第6~10周各治疗组血压与假手术组、模型组比较,差异均有统计学意义（P〈0.01）;氯沙坦钾胶囊组、天麻钩藤颗粒组、超微高剂量组、超微低剂量组与传统剂量组比较,差异均有统计学意义（P〈0.01,P〈0.05）。模型组FBG、FINS、ISI及氯沙坦钾胶囊组FBG、天麻钩藤颗粒组FBG水平与假手术组比较,差异均有统计学意义（P〈0.01,P〈0.05）;风眩宁方超微高、低剂量组及传统剂量组FBG、FINS、ISI水平与模型组比较,差异均有统计学意义（P〈0.05,P〈0.01）;超微高剂量组FBG、ISI水平与传统剂量组比较,差异均有统计学意义（P〈0.05）。结论：风眩宁方可降低高血压病肝阳上亢证模型大鼠的血压,保护靶器官,同时可提高胰岛素抵抗指数,超微风眩宁疗效更佳。Objective： To observe the efficacy of Fengxuanning prescription in the treatment of hypertension and compare the efficacy between Supermicro- Granule and traditional decoction,and to investigate the action mechanisms of the prescription. Methods： A rat model of hypertension with syndrome of upper hyperactivity of liver yang was established by left renal artery stenosis surgery. Sixty rats with hypertension were randomly divided into model group,losartan potassium group,Tianma Gouteng Granule group,high- dose Fengxuanning Supermicro- Granule group,low- dose Fengxuanning Supermicro- Granule group,and traditional- dose Fengxuanning prescription group. Twelve rats were assigned to the sham surgery group. The treatment groups were treated with respective drugs,while the model group and the sham surgery group were given an equal volume of distilled water by gavage. The change in blood pressure was evaluated in each group. The enzymatic method and radioimmunoassay were used to determine the levels of fasting plasma glucose（ FPG）and fasting insulin（ FINS） in rats,respectively. The insulin sensitivity index（ ISI） was calculated. Bilateral renal tissues of the rat were collected for the microscopic morphological analysis. Results： At 6- 10 weeks after model establishment,the treatment groups had significantly lower blood pressure than the model group or the sham surgery group（ P 0. 01）; compared with the traditional- dose Fengxuanning prescription group,the blood pressure was significantly lower in the losartan potassium group,the Tianma Gouteng Granule group,the high- dose Fengxuanning Supermicro- Granule group,and the low- dose Fengxuanning Supermicro- Granule group（ P 0. 01; P 0. 05）. Compared with the sham surgery group,the model group had significantly different levels of FBG,FINS,and ISI,while the losartan potassium group and the Tianma Gouteng granules group had significantly different levels of FBG（ P 0. 01; P 0. 05）.
