摘  要:目的:研究蛇伤药酒的质量控制方法。方法:采用薄层色谱法(TLC)对制剂中大黄进行定性鉴别;采用高效液相色谱法(HPLC)测定盐酸小檗碱的含量,Diamonsil C_(18)(2)色谱柱(200mm×4.6mm,5μm),流动相为乙腈-0.05mol/L磷酸二氢钾溶液(32:68),检测波长为345nm,流速为1.0m L/min,柱温为25℃。结果:定性鉴别薄层色谱斑点特征明显;高效液相色谱法测定盐酸小檗碱含量,在0.04~0.42μg范围内与峰面积值呈良好的线性关系,回归方程:Y=0.6156X-0.0776(r=0.9999),平均加样回收率为99.71%,RSD=1.44%(n=6)。结论:采用TLC法对大黄定性鉴别,HPLC定量测定盐酸小檗碱含量,两方法简便准确,重复性良好,可有效控制该制剂质量。Objective:To study the method of quality control for Sheshang medicinal liquor. Methods: The thin layer chromatography (TLC) was used to the qualitative identification of rheum officinale; the quantitative analysis of berberine hydrochloride was carried out by HPLC with a Diamonsil C,s (2) chromatograhpic column(200mm×4.6mm,5um) ,the mobile phase was acetonitrile - 0.05mol/L potassium dihydrogen phosphate solution (32:68) with the flow rate of 1.0 mL/min and the detection wavelength of 345nm, the tem- perature of column was 25 ℃. Results : The characteristic of TLC spots was obvious in the qualitative identification. HPLC determind the content. The berberine hydrochloride in 0.04 - 0. 42ug extent show good linear ralation, and the regression equation was Y = 0. 6156X - 0.0776, ( r = 0.9999) with an average recovery rate of 99.71% ( RSD = 1.44%, n = 6). Conclusion: The methods of using TLC and HPLC are simple and accurate, and can be used for the quality control of Sheshang medicinal liquor.
