摘  要:以牛血清蛋白等标准蛋白为材料进行PAGE电泳,并对多种染色、脱色方法进行研究比较和改进。结果表明:用考马斯亮蓝R-250微波中火染色5min后,再间隔微波中火脱色3个5min后,可观察实验结果,继续摇床脱色1~2 h后,背景低、灵敏度高;用考马斯亮蓝G-250微波染色后,用水漂洗约30min就可达到粗略观察实验结果的目的,若继续在去离子水中浸泡,则时间越久,背景越清晰;脱色时用乙醇替代甲醇,脱色效果相当。The trail was carried out with the standard protein such as bovine serum albumin by polyacrylamide gel electrophoresis( PAGE) to study and improve staining and destaining methods of protein suitable to educational activities. The results showed that the staining solution with Coomassie brilliant blue R250 in the microwave oven for 5 minutes and the destaining in the microwave oven for 5 minutes three times were considered a new method of rapid staining technique of protein,hours for staining and elution are shortened by this technique. The SDS-PAGE was stained with the Coomassie brilliant blue G250 in the microwave oven,the experimental results was observed after with deionized water for 30 minutes. The background of the gelatin is clearer when was decolorized for long time. Compared with methanol the effect and sensitivity of decolorization with ethanol was comparable.
