摘　　要：在1株污染半夏组培苗的枝状枝孢真菌（Cladosporium cladosporioides DF15）中检测到5条双链RNA（double-strand RNA,dsRNA）条带.通过M-SPAT（modified single-primer amplification technique）、克隆测序获得5条dsRNA的序列信息,分别为dsRNA1（2434bp）、dsRNA2（2241bp）、dsRNA3（2008bp）、dsRNA4（1261bp）和dsRNA5（942bp）.经Blast比对,dsRNA1编码蛋白的氨基酸序列与侵染荚孢壳科、球腔菌科真菌的双分病毒属（Partitivirus）病毒,以及侵染辣椒等植物的双分病毒科（Partitiviridae）病毒的依赖于RNA的RNA聚合酶（RNA-dependent RNA polymerase,RdRp）序列相似度最高.氨基酸序列系统进化树分析表明,dsRNA1与侵染真菌的双分病毒属病毒亲源关系更近.通过核苷酸序列5＇非编码区（untranslated regions,UTR）的比对发现,5条dsRNA 的5＇UTR序列相似度高达50.0%-95.3%,并含有2段保守序列5＇-GUAAAUAAACCUU-3＇和5＇ -C（/G）TA（/AG）CAAGT（/C）AT-3＇.根据上述分析,推测5条dsRNA序列来自同1种病毒基因组,并且为侵染C. cladosporioides 的1种新双分病毒科双分病毒属病毒,命名该病毒为C. cladosporioides virus 1.DsRNA (double-strand RNA) extraction was performed and five dsRNA segments in the contaminated fungi (Cladosporium cladosporioides DF15) from Pinellia tissue culture were found. The five dsRNA segments were obtained with cloned and modified single primer amplification technique (M-SPAT) and were determined dsRNA1 (2434bp), dsRNA2 (2241bp), dsRNA3 (2008), dsRNA4 (1261bp) and dsRNA5 (942bp), respectively. The amino acid sequence encoded by dsRNA1 was similar to the segments of RNA-dependent RNA polymerase (RdRp) with Partitivirus of Sporormiaceae sp. and Mycosphaerellaceae sp., Partitiviridae of plant like pepper though blasting in GenBank. The result of the phylogenetic tree analysis showed dsRNA1 and the viruses in Partitivirus infecting fungal were in the same cluster. The five sequences showed that the similarly between 5'UTR was 50%-95.3%, and the 5'UTR of the five sequences contained two regions 5'-CGUAAAUAAACCUU-3' and 5'-C(/G)TA(/AG) CAAGT(/C)AT-3'. In summary, it can be inferred that the five dsRNAs came from the same virus, and this virus named C. cladosporioides virus 1 was a novel virus of partitivirus infecting C. cladosporioides.
