摘  要:目的 观察牧犬接种鼠疫强毒菌后,牧犬血清F1和IgM抗体变化,探讨F1和IgM抗体在鼠疫监测中的意义。方法5只牧犬,采用背部皮下多点注射法向牧犬接种鼠疫强毒菌,测量肛温检查牧犬体温,酶联免疫吸附试验(ELISA)夹心法检测牧犬血清鼠疫F1抗体,ELISA捕获法检测牧犬血清鼠疫IgM抗体。结果接种鼠疫强毒菌第3天,牧犬血清中检出F1抗体,高峰期为第5~58天,平均滴度为1:2^8.20~1:2^11.60。随后滴度缓慢降低.第560天仍有1只牧犬滴度为1:2^5。接种鼠疫强毒菌后第5天,牧犬血清中检出IgM抗体,高峰期为第5-23天,平均滴度为1:2^8.60~1:2^12.00随后滴度快速降低,第46天以后降到阳性标准以下。结论ELISA夹心法检测牧犬血清鼠疫F1抗体。阳性时间长达560d,可用于追溯动物间鼠疫流行,特别适用于疫源地调查。EUSA捕获法检测牧犬血清鼠疫IgM抗体,阳性时间仅40d,更适用于判断近期动物间鼠疫流行状况。Objective To observe the dynamic trend of serum F1 and IgM antibody of shepherd dog inoculated with virulent Yersinia pestis, and to study the significance of F1 and IgM antibody in plague surveillance. Methods Five shepherd dogs, virulent Yersinia pestis was inoculated by subcutaneous multipoint injections on back of shepherd dog, rectal temperature was taken to represent body temperature, plague F1 antibody was detected by the method of sandwich enzyme-linked immunosorbent assay (ELISA), while plague IgM antibody was detected by capture ELISA. Results Plague F1 antibody was detected after inoculation with virulent Yersinia pestis on the 3rd day, the peak on the 5th day to 58th day and average titer of 1 : 2^8.20 1 : 2^11.60, then, the titer reduced slowly, and on 560th day, there was a dog with the titer of 1 : 25. Plague IgM antibody was detected after inoculation with virulent Yersirria pestis on the 5th day, the peak on the 5th day to 23rd day and average titer of 1 : 2^8.60 1 : 2^12.00, then, the titer decreased rapidly and went down to the positive standard after the 46th day. Conclusions The results of the antibody induced by inoculation with virulent Yersinia pestis have showed that, the positive plague F1 antibody in the dog detected by sandwich ELISA is lasted for 560 days. So it can be used to detect plague prevalence in animals, especially to investigate the epidemic focus. The positive plague IgM antibody detected by the capture ELISA is lasted for only 40 days, and it is suitable for judging recent epidemic situation of plague in animals.
